Objective:Observing Chinese Herbal Compound formula-Ruyanning and Exemestane on tumor weight,to find out if Chinese Herbal Compound formula-Ruyanning could enhance the effect of exemestane inhibiting tumor.Observing Chinese Herbal Compound formula-Ruyanning and Exemestane on expressions of glucose transporter-1(GLUT1)and Pyruvate kinase M2 isoform(PKM2)of breast tumor-bearing nude mice,to reserch the possiblemechanisms of Ruyanning interfering endocrine therapy.Material andmethods:Human breast cancer MDA-MB-435 of logarithmic phase were digestedby trypsin preparing of cell suspension.Then they were injected into axillary subcutaneous tissue of female mice,a total of three,bilateral ovaries removed.After inoculation,the mice were raised in SPF animal laboratory of Liaoning university of Chinese medicine experiment center.Watch the general state of the mice and tumor formation every day.When the tumor tissue was 0.8 cubic centimeter,take out the tumor tissue in the underarmin of the three mice in aseptic conditions and transplant the tumor tissue in underarmin of the 40 nude mice respectively.Continue to raise the micenormally and watch the tumor formation every day.The animal model of breast tumor-bearing nude mice was built successfully in about 15 days and tumor rate was 100%.After the tumor-bearing nude mice model of breast cancer was established,successfully,the mice was divided into control group,Exemestane group,joint group(Ruyanning+Exemestane)and Ruyanning group randomly.Each group has ten mice.According to the human and mouse equivalent dose conversion formula,0.2 ml distilled water was given to mice in control group,0.2 ml exemestane was given to mice in Exemestane group,0.2 ml Ruyanningand Exemestane was given to mice in joint group,0.2 ml Ruyanning was given to mice in Ruyanning group.Every mice wasgavaged once a day for 21 daysin all.After the experiment,all the mice were sacrificed through broken neck death and tumor were weighted afer being taken completely.Then calculate the anti-tumor rate.Tumor tissue were standby.The expression of GLUT1 and PKM2 was detected by using Western Blotting method.Results:1.Tumor weight andanti-tumor rate:The tumor weight(g)of the control group?the Exemestane group?the joint group and the Ruyanning group was 0.723±0.200?0.418±0.227?0.261±0.099 and 0.571±0.078.Among them,the Exemestane group?joint groupand Ruyanning group were all lower than the control group,and there were statistically significant(P < 0.05).The joint group was the lowest,followed by the Exemestane group.Both the joint group and the Exemestane group were lower than the Ruyanning group.There were statistically significant(P<0.05)as well.The anti-tumor rate of joint group,Exemestane group and Ruyanning group was 63.9%?42.2% ? 21.0% respectively.The anti-tumor rate of joint group was highest,and the Exemestane group was higher than the Ruyanning group.2.Expressions of GLUT1 and PKM2:The expressions of GLUT1 of the control group?the Exemestane group?the joint groupandthe Ruyanninggroup was 184.12±3.94?128.44±4.75?106.38±3.3and 155.92±6.68.Among them,Exemestane group?the joint groupand Ruyanning group were all lower than the control group,and there were very statistically significant(P<0.01).The joint group was the lowest,followed by the Exemestane group.Both the joint group and the Exemestane group were lower than the Ruyanning group.There were very statistically significant(P<0.01).The expressions of PKM2 of the control group?the Exemestane group?the joint groupand the Ruyanning group was214.72±7.78?107.38±4.11?91.63±7.68 and 126.19±4.06.Among them,Exemestane group?the joint groupand Ruyanning group were all lower than the control group,and there were very statistically significant(P<0.01).The joint group was the lowest,followed by the Exemestane group.Both the joint group and the Exemestane group were lower than the Ruyanning group.There were very statistically significant(P<0.01).Conclusion:1.Ruyanning can inhibit breast cancer MDA-MB-435 to some extent.2.Ruyanning can enhance the effect of exemestane when they are applied together.3.The possible mechanism of above two kinds of effect is inhibiting tumor cell metabolism by lowing protein expressions of GLUT1 and PKM2.
