PurposeTo observe the effects of Qi-nourishing and blood-activating detoxification on the neointimal hyperplasia and four types of serum inflammation in rat models of restenosis(RS)after restenosis(RS)using percutaneous transluminal angioplasty(PTA).Factors:matrix metalloproteinase-9(MMP-9),interleukin-1(IL-1),interleukin-8(IL-8),high-sensitivity C-reactive protein(Effects of high-sensitivity c-reactive protein(hs-CRP)levels and mRNA and protein expression of calcineurin(Ca2+/CaM-CaN-NFATc)signaling pathway-related factors The molecular biological mechanism of restenosis after stenting.MethodSD rats were randomly divided into sham-operated group,model group,Zhicao Huayu Decoction group,and pravastatin group.Four groups of rats were established.The animal models of balloon-injured rat carotid artery restenosis were established.Blood sampling was used to observe the effects of Fangai Huayu Decoction on the levels of serum inflammatory cytokines hs-CRP,IL-1,IL-8 and MMP-9 in rats with restenosis after PTA.Observe the section of the blood vessels,and observe the intimal hyperplasia after staining.Observe the degree of intimal hyperplasia in each group.Observed by PCR and western blot to observe the effect of Fangai Huayu Decoction on Ca2+/CaM-CaN-NFATc signaling pathway and glycogen synthesis in vascular tissue of restenosis after PTA The effect of mRNA and protein expression of Glycogen synthase-excited(GSK-3B)signaling molecules.By statistical analysis,comparisons between groups were considered P<0.05 to be considered statistically significant.result1 Results of serum inflammatory factors(MMP-9,IL-1,IL-8,hs-CRP)?The serum levels of MMP-9 in rats of the experimental model group were significantly increased,and the contents in the Chinese medicine group were significantly lower than those in the model group(24.36±8.15vs40.13±15.37,*P<0.05).The western medicine group was lower than the model group(23.95±12.15).(vs 40.13±15.37*P<0.05),the content of model group was significantly higher than that of sham group(40.13±15.37vs23.80±8.53,*P<0.05),but the contents of Chinese medicine group and western medicine group were higher than those of sham group.Increased,the other groups were not statistically significant.?Compared with the serum IL-1 level in the four groups,the model group(42.2±l7.21)>sham group(33.79± 13.92)>Western medicine group(33.04±13.33)>Chinese medicine group(26.40±10.55).Compared with the model group,IL-1 in the Chinese medicine group was significantly lower,*P<0.05,and there was no statistical difference between the other groups.?The level of serum IL-8 in sham-operated rats was the highest in the four groups(30.74±12.19).Compared with the three groups with balloon-injured carotid arteries,the content of model group was higher than that of western medicine group(30.43±8.35 vs 29.87±12.15),while the Chinese medicine group has the lowest content(23.01±8.46).No statistical difference was found between the four groups after comparison.?The serum hs-CRP content(0.2(0.20-0.32))was the lowest in the four groups,and the three groups of balloon injury were higher in the model group(1.10(1.10-1.20))than in the western medicine group(4.1.00 ± 0.15),while Chinese Herbal Medicine has the lowest content(0.99±0.15).Between groups,the content of the model group was higher than that of the sham group(*P<0.05);the content of the western medicine group was higher than the sham group(*P<0.05);the content of the Chinese medicine group was higher than the sham group(*P<0.05).There was no statistical difference between the other groups.2 PCR detection results ? The mRNA expression of NFATC1 detected by PCR in this study was expressed by 2-??Ct:Compared with the sham group,the mRNA expression of NFATC1 in the other three groups of vascular tissues increased significantly.The mRNA expression of NFATC1 was highest in the Chinese medicine group.The mRNA expression of NFATC1 was compared between the four groups.The Chinese medicine group was higher than the sham group(P<0.05).There was no statistical difference between the other groups.?The mRNA expression of GSK-3p in the four groups was detected by PCR:sham operation group 2.16±1.46,model group 13.67±17.82,Chinese medicine group 36.64±12.79,western medicine group 9.78±10.62,and the sham operation group had the lowest expression level.,namely:Chinese medicine group>model group>western medicine group>sham operation group.Between groups,the expression of Chinese medicine group was higher than the other three groups,and there was statistical significance.3 Western blot analysis Western blot analysis detected the protein expression level of GSK-3? in the blood vessel wall of the four groups,the highest expression level in the sham group(1.54±0.38),followed by the Chinese medicine group(1.18±0.24),and the lowest in the western medicine group(3.0.98 ± 0.21),the model group(1.02 ± 0.17)was slightly higher than the western medicine group.Through multiple comparisons between the four groups,the Western medicine histone protein expression was significantly lower than the sham group(*P<0.05);the model group histone protein expression was lower than the sham group(*P<0.05);no comparison between the other groups.Statistical differences.4 Pathological section observation of intimal hyperplasia results In this experiment,the mean thickness of the intima-media thickness(I/M)in each group was compared by the segment of the target vascular segment and after staining,and I/M was used to represent the intimal hyperplasia.Intimal hyperplasia was significantly increased in the model group.The I/M in the model group,the TCM group,and the western medicine group was significantly higher than that in the sham operation group(P<0.05),while the I/M in the TCM group was significantly lower than the balloon injury group(P<0.05).Chinese medicine group I/M was significantly lower than western medicine group(P<0.05)ConclusionIt can be seen from this experiment that the prevention and treatment of decoction by activating blood and detoxifying blood circulation can prevent the expression of signal molecules on the calcineurin signaling pathway,reduce the degree of intimal hyperplasia and development process,and reduce the inflammatory factors.The content of hs-CRP,IL-1,IL-8 and MMP-9 reduced the inflammatory response and thus prevented restenosis after PTA.Therefore,the prevention and treatment of restenosis after PCI in patients with Niao Hua Hua Tang decoction under the guidance of qi-invigorating,blood-activating,detoxification-resuding can be clinically applied,which provides a new idea for the treatment of this disease with integrated traditional Chinese and western medicine.
