Lung cancer is the most frequent malignant tumor in world,many patients often have metastases in distant places,bone metastasis is the main complication of lung cancer.Thirty to fourty percent of lung cancer patients will have bone metastasis and most of them are multiple.Remodeling of bone is a dynamic process,the osteoblast and osteoclast have play an important role in this activity.The mechanism of neoplastic bone metastasis is influenced by tumor cell, the osteoblast and osteoclast. The interaction among them lead to disequilibrium of normal remodeling of bone.Ca2+ has a important role in physiology and pathology on neoplastic bone metastasis,CaN/NFAT pathway relate to the bone formation and bone absorption.Calcineurin is the only Ser/ THR protein phosphatase adjusted by Ca2+- calmodulince dependent in cells. Half of CaN located in endochylema,the rest integrate to cytolemma. It has high expression in brain, heart and skeletal muscle,and we also can find it in lung, kidney, uterus and liver,it's suggested that CaN may has great physiological functions.The relationship between CaN and bone metastasis in lung cancer has not been reported up to now.Objective: To investigate the expressions of calcineurin (CaN Aαand CaN Aβ) in lung carcinoma with or without bone metastasis and to analyze the relationship between the expression of the members of CaN family and clinicopathological features. Provide theory basis for chinic therapy of lung cancer with bone metastasis。Methods: The cases of lung cancer come from TangDu hospital of Fourth Military Medical University, the patients are hospitalized in chest surgery for lung cancer from June of 2004 to December of 2008。Total to 89 examples(SqCa: 28;adenocarcinoma:28;SCLC:33).We devided the patients into two groups according to clinic and imageology detection.Group A:the petients who were lung cancer without bone metastasis,49 example.Group B:petients who were lung cancer with bone metastasis,40 example.To identify the expressions of CaN Aαand CaN Aβin lung cancer tissues with or without bone metastasis by immunohistochemistry method. Finally invited two Pathologic doctor to examine the immunohistochemistry slice.To identify the expressions of CaN Aαand CaN Aβin SBC-3,SBC-5 and NCI-H345 cell lines by RT-PCR and Western Blot.,emphasis on their gene and protein level.Result:1. There has differences among the expressions of CaN Aαand Aβin lung squamous carcinoma, adenocarcinoma and small lung cancer tissues with and without bone metastasis,almost higher for 40.6%,42.9%,58.7%. The expressions of CaN Aαand CaN Aβwith bone metastasis was higher than the group without bone metastasis in lung cancer by Mann-Whitney U test(.CaN AαZ=19.176,P=0.000;CaN AβZ=15.248,P=0.000 P<0.05).We can see CaN Aαand CaN Aβhave no difference in three group by Kruskal–Wallis H test(CaN Aαχ2=5.117,P=0.079;CaN Aβχ2=4.883,P=0.094 P>0.05).2. Pass through gray scaleanalysis RT-PCR and Western blot's result,we can see the expressions of CaN Aαand CaN Aβin SBC-5 is higher than SBC-3 and NCI-H345,the expressions of CaN Aαand CaN Aβin SBC-5 is higher than NCI-H345 for 3.61and 6.37 times.Conclusion:The expressions of CaN Aαand CaN Aβin special cells and tissues with bone metastasis are higher than that without bone metastsis, The mechanism of action maybe concerned with many signal transduction pathway in bone metastasis.Extremely to study and research CaN pathway maybe provide rationale for the diagnosis and therapy on bone metastasis of lung cancer.
