PurposeThe aim of this study was to establish a rat model of myocardial ischemia-reperfusion injury,to study the expression level of Toll like receptor 2,toll like receptor 4,nuclear factor-? B,the changes of myocardial injury related markers,the pathological changes of myocardial tissue,and to explore the protective mechanism of Tongxinluo.To provide some new basis for the prevention of coronary heart disease.MethodsMale SD rats were randomly divided into sham-operation group model group and Tongxinluo group(12 rats in each group).Coronary artery ischemia was ligated for 1 hour and reperfusion for 2 hours.In the sham operation group,only the thread was pierced and not ligated.The rats in the Tongxinluo group were perfused with Tongxinluo,and the other two groups were perfused with the same amount of 0.9%sodium chloride for one week.After reperfusion,electrocardiogram(ECG)was detected,TTC staining was performed with HE staining,and transmission electron microscopy was observed.The level of CK-MB,LDH,TNF-?,IL-6 was detected by Elisa.The expression of NF-? B,TLR2 and TLR4 was detected by real-time fluorescence quantitative PCR.Results1 Electrocardiogram(ECG):The St segment increased gradually after the establishment of model in each group,and then decreased gradually.There was no difference of heart rate in three groups(P>0.05).Compared with the model group,increase of St segment depression of ECG in Tongxinluo group(P<0.05).2 TTC staining:heart staining was red in sham-operated group,white changes were seen in anterior wall of myocardium in model group,and myocardial white in Tongxinluo group was less than that in model group.3 HE staining:In sham-operation group,the arrangement of myocardial fibers was regular,the intact structure and normal morphology could be seen.Eosinophilic changes and breakage were observed in the model group,but no obvious changes in bleeding and edema were found in the Tongxinluo group,and the infiltration of inflammatory cells was lighter than that in the model group.4 Transmission electron microscope(TEM):In the sham-operation group,the myocardial arrangement was regular and the structure of mitochondria was approximately complete.In the model group,the myocardial arrangement was disorder,the mitochondrial structure was disorder,the myocardium of Tongxinluo group was orderly,the interstitial was not oedema,and most of the mitochondria were normal.5 The expression of inflammatory factors:Compared with the sham operation group,the serum TNF-a,IL-6 expression in the model group was significantly higher than that in the sham operation group(P<0.01),and the expression of TNF-?,IL-6 in the Tongxinluo group was significantly lower than that in the model group(P<0.01).6 Myocardial enzyme expression:The expression of serum CK-MB,LDH in the model group was significantly higher than that in the sham operation group(P<0.01).Compared with the model group,the serum CK-MB,LDH expression in the Tongxinluo group was significantly lower than that in the model group(P<0.01).7 TLR2?TLR4?NF-? BmRNA expression:compared with sham-operation group,TLR2mRNA level in model group increased significantly(P<0.05),compared with model group,the level of myocardial TLR2mRNA in Tongxinluo group decreased significantly(P<0.01),compared with sham operation group,the level of myocardial TLR2mRNA in Tongxinluo group decreased significantly(P<0.01).Compared with the model group,the level of myocardial TLR4mRNA in the Tongxinluo group decreased significantly(P<0.05).Compared with the sham operation group,the myocardial NF-? BmRNA level in the model group increased significantly(P<0.01).The level of myocardial NF-? BmRNA in Tongxinluo group was significantly lower than that in the model group(P<0.05).Conclusion1 The model of myocardial ischemia reperfusion injury in rats was established by ligating anterior descending coronary artery.2 Tongxinluo can relieve myocardial injury,reduce the expression of TNF-? and IL-6,and decrease the expression of myocardial enzyme CK-MB and LDH.3 Tongxinluo can reduce inflammatory reaction through TLR2/TLR4-NF-? B pathway and improve myocardial ischemia-reperfusion injury.4 Tongxinluo can reduce the area of myocardial ischemia and the pathological changes of myocardial ultrastructure in rats.
