Objective:to study the endurance and resistance exercise on the effect of diabetic myocardial sarcoplasmic reticulum calcium recovery and improve the effect of left ventricular diastolic dysfunction.Methods:60 male SD rats,and randomly divided into control group and module,feeding respectively in normal and high sugar,high fat feed.Seven weeks after building module by intraperitoneal injection of one-time chain of 30 mg/kg urea with cephalosporins(STZ),after 72 h,the fasting blood glucose or greater tendency for 16.7 L and insulin resistance in diabetes group.Since then,the control group were divided into blank control group(C),treadmill exercise control group(CA)and gym exercise control group(CR).Diabetes group were divided into control group(D),diabetes,treadmill exercise groups(DA)and diabetes gym exercise group(DR).After eight weeks of exercise intervention,light electron microscope groups of left ventricular myocardial structure;The fasting blood glucose(FBG),blood lipid,insulin(FINS),insulin resistance index(HOMA IR)and glucose tolerance(AUC);Left ventricular echocardiography,myocardial calcium pump and pressure curve(SERCA2a),phosphoprotein(PLB)and cytoplasm,sarcoplasmic reticulum Ca2 +.Results:1.7 weeks high-sugar high-fat feed joint one STZ injection,Ⅱ diabetes rats ChengMo rate was 72.2%;2.Compared with group C,group D cardiac muscle fibril fracture,disorganized,and mitochondria swelling degeneration phenomenon;FBG,triglyceride(TG)and low density lipoprotein(LDL)increased significantly;Increase significantly AUC,significantly lower Fins;Left heart indoor pressure maximum decline rate(dp/dt Max)decreases significantly,the left ventricular relaxation time constant(Tau)significantly longer;SERCA2a,PLB and sarcoplasmic reticulum calcium/cytoplasm fluorescence value was lower than that in group C,sarcoplasmic reticulum calcium distribution reduction and increase in cytoplasm.3.DA ultrastructure is D group improved,but DR group tends to increase;Endurance exercise,TG,LDL drop above resistance movement,and the resistance movement to improve the FBG,FINS and AUC is more obvious;Endurance sports can obviously increase the dp/dt Max,shorten the Tau,the resistance movement-dp/dt Max extent,prolong the Tau;DA SERCA2a protein expression is D group has increased significantly;DR SERCA2a,PLB protein and DA group PLB protein expression have improved,but there was no significant difference in the D group;DA group(sarcoplasmic reticulum calcium/cytoplasm)fluorescence value and DR group increased significantly compared with group D;CA,CR group structure,function and blood indexes,protein expression and sarcoplasmic reticulum calcium/cytoplasm fluorescence value and C group were no significant difference.Conclusion:1.7 weeks high-sugar high-fat feed joint one-time intraperitoneal injection of low-dose STZ copy II diabetes model rats success rate is 72.2%;2.8 weeks course of diabetic rats myocardial sarcoplasmic reticulum calcium recycling and diastolic dysfunction.3.Eight weeks of endurance and resistance exercise can significantly reduce diabetic rats blood glucose levels,improve insulin resistance;In improving blood lipids and myocardial ultrastructure,ventricular diastolic function and calcium regulatory proteins on endurance exercise is superior to the resistance movement;Resistance movement in the lower blood sugar,reduce insulin resistance than endurance exercise is more obvious. |
