According to the existing literature Castanopsis composition are rich in polyphenols.And there are rich in triterpene hexahydroxydiphenoyl esters compound that have a chemical taxonomic significance characteristic.Currently such compounds are only isolated from three kinds of Castanopsis.Whether other Castanopsis containing such novel unique compound worth the wait.Currently on the chemical composition of Castanopsis reported very little.In order to enrich the chemical constituents of the Castanopsis and further rational development Castanopsis plant resources.In this study,Castanopsis fargesii are experimental material about chemical constituents.Content of polyphenols from the seeds of Cyclobalanopsis glauca,Cyclobalanopsis fleuryi,Castanopsis fissa,Castanopsis fargesii and Lithocarpus henry were determined.Its extract antioxidant capacities were compared.Method(1)Isolation and identification of chemical constituents from Castanopsis fargesii:5.2 kg of fresh C.fargesii leaves were chopped thensoaked at room temperature.36.4 L 80%ethanol was added(1:7 by volume of solid to liquid ratio).Extracted twice,each time 6 d.After the extract was concentrated under reduced pressure there is precipitation.The precipitate was collected by filtration.The filtrate was continued to concentrated under reduced pressure to give alcohol-free flavored extract.The precipitateand extract was dissolved.After Sephadex LH-20,MCI gel CHP20P,C-18,HP-20SS column chromatography repeatedly and semi-preparative,preparative high performance liquid chromatography,high-speed countercurrent and other modern separation means to give compound.The use of 1H-NMR,13C-NMR,HMBC,HSQC and other nuclear magnetic resonance spectroscopy,high resolution mass spectral and data analysis structures were elucidated.(2)Polyphenol determination and oxidation resistance of 5 kinds Fagaceae seed:Ripe seeds vacuum dried and crushed then ultrasonic extraction three times.The combined filtrate volume to give the sample solution.Gallic acid as a standard to establish a standard curve regression equation,measuring the sample absorbance at 765nm.Polyphenol content of samples obtained from the regression equation.Antioxidant experiment by DPPH·and ABTS · method.Result:(1)Extract of C.fargesii fresh leaves was isolated 24 compounds including phenolic acids,quinic acids,glycosides and other compounds.Phenolic acids:shikimic acid(J1)?3,4-dihydroxybenzoic acid(J2)?caffeic acid(J3)?gallic acid(J4)?methyl gallate(J5)?ethyl gallate(J6);quinic acids:5-O-galloylquinic acid(J7)?5-O-caffeoylquinic acid(J8)?methyl 5-O-galloylquinic acid(J9);gallic tannins:3-O-galloyl-shikimic acid(J10)?5-O-galloyl-(-)-shikimic acid(J11)?5-O-galloylhamamelose(J12);glycosides:1-O-galloyl-?-D-glucose(J13)?6-O-galloyl-glucose(J14)?1,6-di-O-galloyl-?-D-glucose(J15)?methyl?-D-glucopyranoside(J16)?ethyl glucopyranoside(J17);other compounds:crenatin(J18)?ellagic acid(J19)?((2R,3S,5R,6S)-3,4,5-trihydroxy-6-((2,3,4,5,6-pentahydroxycyclohexyl)oxy)tetrahydro-2H-pyran-2-yl)methyl 3,4,5-trihydroxybenzoate(J20);new compounds:3-hydroxy-3-(1-hydroxyethyl 1-O-?-D-(6',-O-galloyl))dihydrofuran-2(3H)-one(J21)?(3S,4S)3-hydroxymethyl-3,4-dihydro-5,6,7-trihydroxy-4-(4'-hydroxy-3'-methoxyphenyl)-1H-[2]-benzopyran-1-one(J22)?(2S,3R,5S,6R)-methyl 2-(4-((1,2-dihydroxyethoxy)carbonyl)-2,6-dihydroxyphenoxy)-4,5-dihydroxy-6-(hydroxymethyl)tetrahydro-2H-pyran-3-carboxylate(J23)?5-O-(?-rhamnopyranosyl-(1?6)-O-?-glucopyranosyl)gentisic(J24).(2)The highest polyphenols content of the 5 kinds Fagaceae seeds is 14.16%of C.fleuryi followed by C.fissa seed 10.89%.C.fargesii seeds contain only the minimum 0.44%.DPPH ·scavenging composition of C.glauca,C.fleuryi seed extract is quite to Vc.C.fissa seed extract DPPH · scavenging composition is the weakest.The strongest ABTS · clear was C.glauca.In the same concentration C.glauca seed extract IC50better than Vc.C.fleuryi IC50 value is quite to Vc.Lithocarpus henry seed extract ABTS·clear was the weakest.Conclusion:24 Compounds isolated from C.fargesii leaves.All compounds are isolated from C.fargesii leaves first.The four new compounds named:J21,J22,J23,J24.Fagaceae coat is rich in polyphenols.Chemical composition of seeds is starch-based.The seed coat of different plants is so different that polyphenol content among the seeds are quite different.Polyphenol content have certain dose-effect relationship between antioxidant capacity.But the seed extract of greatest polyphenol content antioxidant capacity is not necessarily the best.This may be concerned to polyphenol ingredient categories of plant.
