Cell culture in vitro is a method for the growth and propagation of the cells in the physiological environment,and it is an important technology in the research of biological engineering.Cultured cells can be used to isolate and purify the virus,to produce vaccines and drugs.The success of in vitro cell culture has created a condition for the study of cell structure and function,including the establishment of a variety of cell lines.These cultured cells have become an important way for the study of cell biology,immunology and so on.Cell culture in vitro need to meet some basic conditions: 1.A sterile non-toxic culture environment;2.Suitable culture temperature;3.Suitable gas environment and p H;4.Adequate nutrition,including a variety of amino acids,vitamins,coenzymes,nucleic acids,purines,pyrimidine,growth factors.Swine testis cells(ST cells)are widely used in the breeding of classical swine fever virus(CSFV),porcine parvovirus(PPV),pseudorabies virus(PRV),porcine transmissible gastroenteritis virus(TGEV),etc.,it is one of the important cell lines for virus breeding and vaccine production.In this study,we studied the scale culture of ST cells,by comparing the effects of different serum concentration(5%?10%?15%),different culture temperature(35??36??37?)and different p H(7.2?7.3?7.4)on the culture of ST cells in vitro,and through the cell count method,plotted the growth curves of ST cells cultured under different serum concentrations,different culture temperatures and different p H values,so as to screen out a most suitable for ST cells in large scale in vitro culture process.Experiment shows that,The serum concentration of 10% was the most suitable and the most economical concentration in the serum concentration of three;Three different temperature and p H culture environment had little effect on ST cell culture in vitro.The appropriate serum co ncentration can meet the needs of cell growth,but also to avoid the excess nutrients caused by high serum concentration,and thus produce toxic effects on cells.The results of this study provide a suitable reference value for the serum concentration,temperature and p H required for large-scale ST cell culture in vitro,it is of great significance to improve the production process of swine testis cells to produce swine fever attenuated vaccine.
