With the flourishing development of shipping economy,the frequent occurrence of oil spill accidents followed on.Oil spills have exerted extreme influence on marine environment and subsistence of human beings.In order to clean up and soak up the oil pollution,the following three methods are usually applied: Physical method,Chemical method and Bioremediation method.Through comparison,the Bioremediation method is more useful to deal with the dispersive and dissolved oil pollutions which are difficult to solve by using Physical method,and compare to the Chemical method,it takes the advantages of safety,economy and no secondary pollution,so it catches many scientific and technical workers' eyes.However,the complexity of marine environment severely affects the regular metabolism of microorganism.Thus,using Bioremediation method to deal with oil spills still cannot be applied in reality.In this thesis,the author firstly separated high-efficient petroleum hydrocarbon degrading bacterial from the environment polluted by petroleum hydrocarbon,then approached compound composition experiment and orthogonal experiment,and created multiple flora of high-efficient petroleum hydrocarbon degrading bacterial.Later on,the author made use of bacterial resistance to further improve the technology for adapting the degrading bacterial to the marine environment,expecting to offer a new way of thinking that using Bioremediation method to deal with the oil spills.The results are showed below:Six strains of petroleum hydrocarbon degrading bacteria with good degradation efficiency were selected from the screening of petroleum hydrocarbon pollutants in shuangdao bay beach,the degradation rate of petroleum hydrocarbons within 7 days was between 25.7 and 40.6% by ultraviolet spectrophotometry.Five of the strains with good growth state were selected for the construction experiment of petroleum-hydrocarbon degrading bacteria,the high efficiency oil hydrocarbon degrading bacteria group LQ1 was constructed by complex orthogonal experiment,7d degradation rate reached 65.8% under laboratory conditions.Then,the retrograde improvement was carried out by exogenously adding betaine and immobilizing bacteria,formation of high efficiency complex bacteria for hydrocarbon degradation.Finally,a study on the degradation kinetics of the bacteria found that,bacteria can degrade petroleum hydrocarbons,and with anti-retroviral assistance,the degradation rate of Marine environment has been improved obviously.This research provided a new way of thinking that to deal with oil spills pollution using Bioremediation method.The research achievement immobilized bacteria has a wide application prospect in dealing with oil spills using Bioremediation method.
