Cotton is a drought resistant crop,but the drought problem is still one of the important factors restricting the development of cotton cultivation.In response to the increasingly serious drought disaster,17 cotton varieties were used as test materials to control water stress in cotton blossing and boll-forming stages in this study.The comprehensive drought resistance of 17 cotton resource materials under different traits was evaluated by comprehensive drought resistance coefficient(CDC)? correlation analysis method and D value evaluation method.Then the optimal single index of drought resistance was found out,and the cotton varieties with strong drought resistance were screened out.Based on the early stage of the laboratory through proteomic analysis getting losts of differential expression of protein spots,selected three significantly correlated with drought stress protein samples from them,the full-length cDNA of these three differential protein samples was cloned by homologous cloning,named GhPR-10?GhRBCO and GhSAMS,using the relevant bioinformatics software to predict and analyze their structure and function.The qPCR technique was used to detect the expression of three genes under three non-biological duress about different stress time or different tissue organs.And the plant expression vector of GhPR-10 gene was constructed by using the cloned GhPR-10 gene and pCAMBIA1304 plasmid as plant expression vector and binding primers with NcoI and SpeI restriction sites.The main research results of this study are as follows:1.As the D value can not only reflect the drought resistance of each index,but also reflect to the contribution rate of drought resistance to the size of the variety,and can be more comprehensive response to the variety of drought resistance size,so unified D value system cluster analysis of 17 cotton resource materials.Therefore,the D value system cluster analysis is used to classify the drought resistance of resource materials.D-value clustering results were different under different indexes.Based on the results of D value evaluation of the photosynthetic indexes of drought resistant type materials mainly include Lumian 28?Shinong 98-7?10615-1?Junmian 1 and KK1543 and so on;based on the results of D value evaluation of fiber quality indicators drought resistance type materials mainly include ZhongR 2007?Lumian 28?CQJ-5?Keyang 1?KK1543?Xinluzao 36?Xinluzao 47 and XinshiK 7;based on the results of D value evaluation of agronomic indexes drought resistance materials mainly include Junmian 1?XinshiK 7?CQJ-5?Xinluzao 36 and KK1543 and so on.According to the results of D value evaluation of three traits,KK1543 varieties were selected as follow-up test materials in combination with the characteristics of suitable indoor water culture seedlings.2.The predicted results indicate that the proteins encoded by GhPR-10?GhRBCO and GhSAMS are hydrophilic and do not have the signal peptide and transmembrane structure.Subcellular localization prediction showed that GhPR-10 and GhSAMS were localized in the cytoplasm,while GhRBCO was located in the chloroplast.3.The qPCR results showed that GhPR-10?GhRBCO and GhSAMS were up-regulated in drought?low temperature and salt stress,all of which could respond to stress,but the expression pattern was significantly different.Under the drought and salt stress,GhPR-10?GhRBCO and GhSAMS in the roots?stems and leaves of cotton were all tissue-specific.The plant expression vector pCAMBIA1304-PR-10 of GhPR-10 gene was constructed,which will lay a foundation for further study of GhPR-10 gene function.
