Urinary tract infections(UTIs)is a common disease of lower urinary tract syndrome in pets.uropathogenic E.coli(UPEC)is a common pathogen of UTIs.UPEC has not only a huge harm to pets,but also the possibility of infecting people through pets.In China,the study of UPEC is still in the primary stage.Currently,the research is mainly focused on UPEC in human,while lacking of these in pets.The relevant epidemiological data is not perfect.In this study,drug sensitivity,biofilm phenotype,phylogenetic groups,virulence genes and drug resistance genes of UPEC strains from canines and cats between Taiwan and Jiangsu regions were detected and compared.The research results will lay the foundation for further study of the pathogenesis,drug resistance mechanism,and prevention and treatment of UPEC.Antibiotic sensitive test were performed on 76 UPEC strains from Taiwan(n=59)and Jiangsu(n=17).The results showed that strains have produced varying degrees of resistance to most antibiotics,and 80.26%of the strains were resistant to amoxicillin,following by 72.37%to mezlocillin.The rate of multiple drug resistance was 75%.The most sensitive drug was cefoperazone with a sensitivity rate of 98.68%.The UPEC strains isolated from Taiwan(n=59)showed resistant to up to 13 antibiotics,and the resistance rate to amoxicillin was as high as 81.36%,however,all of them were sensitive to cefoperazone.The UPEC isolates in Jiangsu(n=17)were resistant to 12 antibiotics,and the resistance rate to mezlocillin and amoxicillin are as high as 76.47%.The sensitivity rate of cefoperazone was about 94.12%.The data analysis showed that there were significant differences in drug resistance rates between the UPEC of these two regions,such as tobramycin,amikacin,neomycin,ceftriaxone,doxycycline and polymyxin(P<0.05).The biofilm phenotype of 76 strains of UPEC was determined by semi-quantitative determination of 96 orifice plate and microscopic observation.The results showed that the biofilm-forming rate of UPEC strains was about 76.32%(58/76),which was mainly strong biofilm formation ability,accounting for about 36.84%.The biofilm-forming rate of UPEC in Taiwan is 71.19%(42/59),which mainly exhibited the strong biofilm formation ability,accounting for about 33.90%.The biofilm-forming rate of UPEC in Jiangsu is 94.12(16/17),which mainly showed the strong biofilm formation ability,accounting for about 36.84%.The phylogenetic groups,18 virulence genes and 15 drug-resistant genes of 76 UPEC strains in canines and cats from Taiwan and Jiangsu regions in China were detected and analyzed by PCR method,respectively.Results of phylogenetic analysis showed that phylogenetic groups in UPEC were mainly group B2(30.26%)and group D(27.63%).The strains isolated from Taiwan were mainly group B2(27.12%)and group D(32.2%),and the isolates in Jiangsu were mainly group B2(41.18%)and group A(35.29%).Results of virulence genes showed that of the 18 virulence genes examined,fimH carrying rate was 98.68%(75/76),and 31.58%(24/76)strains were found to carry P pilin-related genes,of which the carrying rate of papC gene was 25%(19/76),and no papG allele I and papG allele I' and cdtI genes were found.Among the strains in Taiwan,the carrying rate of fimH was 98.31%(58/59),and the carrying rate of the remaining virulence genes was less than 48%.However,the carrying rate of fimH was 100%(17/17)in Jiangsu,and the carrying rate of the remaining virulence genes ranged from 5%to 42%.There were significant differences in the carrying rates of iut in UPEC strains btween the two regions(P<0.05).Results of drug-resistant genes showed that of the 15 drug-resistant genes examined,the carrying rate of sull was 90.79%(69/76),followed by 69.74%for TEM(53/76),and no CTX-M-2 was found.The sull gene carrying rate of strains in Taiwan was 93.22%(55/59),and no CTX-M-2,SHV and armA were found.Additionally,the carrying rate of the remaining drug-resistant genes was 3%to 70%.The sull gene carrying rate in strains from Jiangsu was 82.35%(14/17),no CTX-M-2 was found.And the carrying rate of the remaining drug-resistant genes was 5%to 71%.There were significant differences in the carrying rates of CTX-M-1,SHV and sul3 in UPEC strains between the two regions(P<0.05).In addition,statistical analysis showed that for the virulence genes,the number of virulence genes carried by UPEC in group B2 was higher than that of groups A,B1 and D.There were significant differences in the carrying rate of virulence genes papA,papC,papG allele ?,focA,focG,sfaS,sat,cnfl,hlyD,ireA,iroN(P<0.05)among phylogenetic groups.For the drug resistance gene,the number of resistant genes carried by UPEC in group B1 was higher than groups A,B2 and D,and there were significant differences in the carrying rate of drug-resistant genes SHV,aac3'-?,qnrD in phylogenetic groups.There was a correlation between biofilm phenotypes of UPEC and drug-resistant genes(aac6'-IB,qnrD,sul3,tetc)(P<0.05).Also,there is a correlation between biofilm phenotypes of UPEC in Taiwan and the resistance of levofloxacin,streptomycin,Polymyxin(P<0.05),there is a correlation between biofilm phenotypes of UPEC in Jiangsu and the resistance of kanamycin.
