Biochip or bioarray,integrate the chemistry and biological response on a single chip according to the Specific functions between different kinds of Biological molecules,to detect the DNA,RNA,Polypeptide,proteins and other kinds of biology elements at a High throughput.PCR chip is one of the biochips based on PCR principle to amplify DNA at a high speed,and it's developing quickly with a wide use range.Micro-Electro-Mechanical System,which is called MEMS for short,is an Emerging technology based on the Semiconductor technology,and it mixes the film,Lithography,LIGA,corrosion,silicon and Non-silicon Processing,and accurate machining technology to produce Electronic components and sensors.If we combine the MEMS technology and biochip together,then we can miniaturize the traditional PCR instruments to a small scale with keeping its DNAAmplification functions.In this paper,we will optimize the structure,decrease the Complexity of the processing to get new type PCR chip with high function Efficiency and simple operation,besides,we will integrate the temperature controlling circuit to build the PCR platform.Our Achievements are listed as following:1.We optimize the PCR chip structure,including choosing new material,redesigning the whole distribution;altering the processing for the chip,including redesigning the mask,reducing the cover made of Polymer,optimizing the glass base and designing new packing technology.Finally,we got a Rectangle Micro fluid chip,18x12mm~2,which has a simple structure and more Uniform temperature Distribution and better Sealing property.2.Build temperature-controlling module,and combine it with the fluid micro chip above,resulting in the whole PCR chip platform.In this paper,we employed labVIEW to control the input and output of the Data acquisition,and combine it with the outer circuit to drive the temperature change to finish the reaction.In our design,the outer circuit included source,the Collection and Amplifier of voltage,heating and cooling modules to fit the need of the temperature.3.We analysis the factors leading to the failure of the PCR chip,and gather these information to get a reliable and stable chip with easy Sample injection and package processing,besides we Eliminated the bubbles forming in traditional PCR chip and improve the stability and uniformity of the temperature.Finally,we use 5 different templates and corresponding Primer to test the effect of the chip and compare the results with traditional PCR instruments.
