Micro-RNA(miRNA)was discovered in 1993.It is a kind of endogenous non-coding single stranded RNA between 18~23nt in various animals,plants and viruses.miRNA through the regulation of post transcriptional gene expression,involved in each process control organisms,covering the growth,differentiation,metabolism,apoptosis,disease process,its expression plays a crucial role in the gene regulation pathway.Pardosa pseudoannulata is an significant species in all spiders in South China rice fields.There are many characteristics,such as large size,wide ecological distribution,strong predation ability and so on in Pardosa pseudoannulata.It plays a momentous role in pest control in rice growing areas in China.In this study,bio-informatics analysis was used to analyze the results of miRNA sequencing of Solexa platform to study the expression level of miRNA and its target genes' function in the ovary of the spider.Specific results are as follows:EdgeR software was used to analyze the differential expression of conservative and newly excavated miRNA in the ovary.A total of 41 miRNA were found differentially expressed in 4 samples,of which 40 were known to be miRNA gene and 1 were unknown miRNA genes.MiRNA was selected from the differentially expressed miRNA to predict the target gene.A total of 64 EST sequences were predicted as possible miRNA target genes.The miRNA target genes of up regulation and down regulation were analyzed by Blast2 go software,and the results showed that miRNA target gene KEGG analysis results were enriched in MAPK signaling pathway and PI3K/Akt signaling pathway related loci.Based on the existing related research,the expression of pseudoannulata in ovary and yolk miRNA does not directly regulate protein synthesis related genes,but through the PI3K-Akt signal pathway affected the synthesis of yolk protein;at the same time,MAPK signal pathway affected the oocyte meiotic division potentially,result in the rate of egg sac.
