With the dramatically increasing awareness that probiotics can benefit the human in numerous ways such as regulating the intestinal environment and maintaining the balance of the flora in the intestine,probiotics is applied in a multitude of industries in the worldwide as examples of the food and the medical sectors.However,some restrictions,such as the low acid resistance and the unendurable storage,may cause the inadequate performance of probiotic count after entering into the human bodies.Specifically,This study mainly focus on how to increase the survival probiotic count in the intestines to achieve the most effective level.In detail,the prodiotic count was tested in the condition of both lactobacillus acidophilus and bifidobacterium separately and compound,meanwhile,some oligosaccharide fructose was added because it has antioxidant polyphenols and can stimulate the growth of probiotics?Besides,plenty of methods were applied to increase the survived prodiotic count in the intestines,such as the Microcapsule technology.1.The study examined the growth curve of the mixed bacteria with the proportion of lactobacillus acidophilus and bifidobacterium being1:1 and get the conclusion that 20 hours after the vaccination is the ideal time with the most prodiotic counts.Next,a single factor test was applied to test how different level of concentration among oligofructose,grape polyphenols and tea polyphenols would influence the probiotics.At the same time,based on the analysis of the optimal combination among those three parameters above,this report summed up the optimal prodiotic count would be 7.65×10~9cfu/mL.when the percentage of oligofructose,grape polyphenols and tea polyphenols is 1.50%,0.080%and 0.080%respectively.2.The optimum conditions for the preparation of probiotic microcapsules by emulsification method were studied.The enzymatic rate of probiotics and the number of viable bacteria after embedding were used as indicators.The single factor experiment was used to systematically explore sodium alginate and carbonic acid.The effect of calcium mass ratio,bacterial-to-gel ratio,water-oil volume ratio,stirring speed,emulsifier addition amount and emulsification time on probiotic microcapsules.After the single factor experiment,orthogonal experiments were carried out on four factors that had a great influence on the embedding rate and viable count.The four factors were the ratio of bacteria to gel,the ratio of sodium alginate and calcium carbonate,and the amount of glacial acetic acid.And stirring speed.The optimum process conditions were as follows:the mass ratio of sodium alginate to calcium carbonate was 1:4,the ratio of bacteria to rubber was 1:4,the stirring speed was 600rpm,and the amount of emulsifier added was 1.6 ml.At this time,the embedding rate measured by the experiment 87.4%,and the viable cell count was 7.65×10~9cfu/mL The microcapsules have good shape,high mechanical strength and no adhesion.3.Explore the storage resistance,gastric acid resistance and enteric solubility of probiotic microcapsules.After storage foSr 60 days at 0?the survival rates of the three samples were as follows:1:1 mixed double-microcapsules(83.5%)supplemented with oligofructose,tea polyphenols,grape polyphenols>unadded mixed double bacteria micro Capsules(80.3%)>mixed double bacteria(62%).After storage for 60 days at 37°C,the survival rates of the three samples were as follows:1:1 mixed double-microcapsules(77%)supplemented with oligofructose,tea polyphenols,grape polyphenols>unadded mixed double bacteria micro Capsules(70%)>Mixed double bacteria(6%).After 2 hours of simulated artificial gastric juice,the survival rates of the three samples were as follows:1:1 mixed double-microcapsules(78.3%)with oligofructose,tea polyphenols and grape polyphenols added>unmixed double bacteria Microcapsules(75%)>mixed double bacteria(2%).In artificial intestinal juice,the microcapsules can be successfully disintegrated within 30 minutes.At this time,no solids exist.
