MicroRNAs are small,non-coding RNAs that can have complement binding to special mRNAs to regulate the target genes expression by transcription repression.In the functional study of microRNAs,target genes can be predicted by bioinformatics methods.However,there are so many related algorithms and tools,it is urgent to find a solution for researchers to make reasonable choices digging out comprehensive and effective information.For this situation,the thesis aims on designing and implementing a microRNA target genes search system,both in more stably updating and credible prediction or verification tools,and in data mining of more visual and clear results.This sustainable,flexible microRNA target genes search system was designed and implemented with visual network and statistical sorting results,using microRNA target genes information extracted from network sources of several commonly used prediction or verification databases,conforming the raw data of different tools to acceptable united formats and applying normalization to them for adjusts core which is the basic score for confidence detecting.The whole system consists of a raw database,a search database creating tool and a series of scripts for searching output results.For sustaining updating and flexibly adding new tools,the raw database contains the raw data from different prediction or verification tools without modification.Data Normalization was done by the search databases creating tool which also called adjust score updating tool,adjust score is the confidence score for statistical sorting.The adjust scores were calculated while new search database was created following 5 normalization steps with box-cox and scale algorithm,in the end a SQL file was generated.The series of scripts were related with preparing,database searching,statistical counting,result producing and network producing.After using the SQL file to rebuild the search database,users providing a microRNA list and a proper configuration file,can easily get summary information of querying microRNAs' target genes,statistical counting and a network diagram showing core results with flexible scale setting by users.Finally,the functional testing using two standard configuration settings of strict and default ones,different tools setting patterns from mixed ones,with score ones to without score ones,and tools number from 1,3 to whole,succeed processing 24 search examples.In the other way,prediction information of the mir-17/92 cluster target genes were searched and results were generated,and the difference between the similar integration system on web such as mirwalk2.0,comiR and the search system of this thesis are compared,and it proves that the results of the system is credible,as well as convenient,flexible and visual.
