Rabies is a zoonotic disease caused by rabies virus(RV)and is a clinical manifestation of acute progressive encephalomyelitis.The fatality rate of rabies is 100%,but there is no effective treatment to date.Rabies is popular in economically underdeveloped areas,and it is reported that about 55000 people all over the world die from the disease every year,most of them children.China is also one of the rabies epidemic areas,with an average annual incidence of more than 1500 cases.About half of them occur in Guangxi,Hunan and Guizhou provinces.In these three provinces,dogs carry RV rate up to 2.3%.At present,the prevention of rabies is still the main reason.Once there are obvious clinical symptoms,even if it is treated with classic Milwaukee protocol,there are few survival cases.Therefore,it is urgent to explore effective treatment.Although the rabies virus infected naturally and immunological histochemical staining of animal models through peripheral infection show that rabies virus is widespread and abundant in the central nervous system,the neuropathological changes caused by rabies virus degeneration are not obvious.At present the treatment of rabies means a single,the treatment effect is not ideal,because of the rabies virus reside mainly in the hippocampus,so this experiment of magnetic nano carrier equipped with anti RV monoclonal antibody to bypass the blood-brain barrier into the effective way of hippocampus to explore,explore more effective way for the treatment of rabies immune therapy.The main contents include: preparation and identification of a gold magnetic nano carrier;the purity of two,anti RV monoclonal antibody and coupling with GoldMag nanoparticles;three,the two routes of administration after administration of rat brain tissue sections immunohistochemical analysis;four,the application of Western Blot technology,content anti AV antibodies were examined in rat brain tissue;DTI imaging,five rat brain.The particle size of the nanoscale carrier is about 80-100 nanometers,and it is uniform in shape and meets the requirements of nanoscale.The purity of RV monoclonal antibody was high,the molecular weight of heavy chain was in 55 KDa,and the molecular weight of light chain was about 30 KDa.The results of immunohistochemistry showed that the number of positive granulocytes in the brain tissue of rats injected with nanocomposite antibody through carotid artery was far more than that of the positive granulocytes in the brain tissue of rats injected with nanocomposite antibody,about two times.But the number of positive cells with simple injection of antibodies is very small,which can basically determine the inantibody to the brain.Western Blot results showed that the content of anti AV monoclonal antibody in rat brain tissue was also more than that of the rats treated with nasal spray after the injection of the carotid artery.The results supported the results of immunohistochemistry.The results of DTI in rat brain showed that the DTI parameters changed before and after administration,and there was a significant difference between them(P<0.05),indicating that anti RV antibody entered the rat brain.According to the data obtained,gold nanoparticles can carry rabies virus monoclonal antibody through the blood-brain barrier and reach the hippocampus of the rat brain,which can become one of the ways to treat rabies.
