Caulerpa lentillifera was edible green algae belongs to Chlorophyta,Bryopsidales,Caulerpaceae.In this paper,polysaccharides were extracted from Caulerpa lentillifera collected in Vietnam.The structures of the polysacgharides were elucidated on the basis of chemical and modern spectroscopic analysis.The bioactivities of the polysaccahrides were also investigated.The main results were as follows:The polysaccharides from Caulerpa lentillifera were extracted by ultra-pure water at80?for 3 hours and precipitated by 75%ethanol at 4?.The yields of the polysaccharides were 5.04%.The polysaccharide fractions CP0-1 was purified and obtained by Q Sepharose Fast Flow ion-exchange and Sephacryl S-200 gel-permeation chromatography.The physicochemical analysis showed that the total sugar content of the CP0-1 was 61.1%,protein was 9.5%,sulfate ester was 11%.The monosaccharide composition showed that CP0-1 was mainly composed of mannose,galactose and xylose at the ratio of 4.4:4.0:1.4.It indicated that CP0-1 was a sulfated xylogalactomannan.The average molecular weight of CP0-1 by gel permeation chromatography was estimated to be about 471 kDa.The structure of CP0-1 was analyzed by degradation,desulfation and methylation.The results showed CP0-1 was was mainly composed of?4)-Manp-?1?as the main chain and the branch at C-3 possibly.Sulfate groups were substiuted at C-3 of?2?-Manp-?1?.Galactose was at the branches with the linkages of?3?-Galp-(1?and terminal Galp-?1?,The sulfate groups were modifiedn at the C-6 of?3?-Galp-?1?.Xylose was located at the branches with terminal and?2?-Xylp-?1?linkaged,there were sulfate modification at C-4of?2?-Xylp-(1?.Degradation of CP0-1 by H2O2 at different temperatures obtained 8 components with different molecular weights.The anticoagulant activities of CP0-1 and degradation components were studied by APTT,TT and PT.CP0-1 displayed intrinsic anticoagulant activity.After mild degradation,CP0-1-2 with molecular weight at 230 kDa displayed more obvious anticoagulant activity.In vitro and higher binding affinity to AT?than CP0-1.Then it achieve anticoagulant and antithrombotic effect through the inhibition the activities of F?a and F?a.The immunoregulatory activity of CP0-1 showed it has a good intestinal immunoregulation effect,which could repair the immunosuppression by cyclophosphamide.And It has the ability to improve the antioxidant level by increasing the activity of antioxidant enzymes in the body.CP0-1 also possessed high potent anti-HSV1 activity by the antivirus ability screening.The results of this paper is of great significance to the utilization and development of the green algae Caulerpa lentillifera.It provided reference on the research of Caulerpa lentillifera.
