The corn germ oil deodorization distillate is rich in squalene,free fatty acids,vitamin E,phytosterol and phytosterol ester,which is the source of vitamin E and phytosterol.The extraction,separation and determination of phytosterol and vitamin E from corn germ oil deodorization distillate,and functional nature of phytosterol was investigated in this paper.The crystallization and recrystallization methods for separation and purification of phytosterol were sdudied,the results show that the opitimum conditions were using acetone-methanol?4:1,V/V?mixed solvent,liquid to material ratio of 3:1,the crystallization temperature of-4?,crystallization time 12 h,acetone as the best recrystallization solvent,the yield of sterol was 70%,and the purity was up to 83.2%;And by using high performance liquid chromatography?HPLC?analysis method to determine the composition of the extract of phytosterol,the composition is campesterol,stigmasterol and?-sitosterol,the ratio of 1:7:6.The extraction of vitamin E by saponification were investigated,and through the single factor and orthogonal test to determine the optimum conditions for extraction vitamin E,the alkaline to material ratio?m:m?of 3.5%,saponification time 4 h,saponification temperature of 55?,the yield of the vitamin E was54.5%under the conditions.The electrochemical reaction process of plant sterol in the bare glassy carbon electrode was investigated in this paper,a new method was developed for the differential pulse rapid determination of plant sterol in oils and food.Experiments showed that the plant sterol appeared oxidation peak at 1.72 V,no reduction peak by fast cyclic voltammetry determination;The electrochemical conditions were optimized to determine phytosterol,theresultsshowedthattheoptimalorganicsolventwas acetonitrile-2-propanol?9:1,V/V?,supporting electrolyte was 50 mmol/L LiClO4,pulse amplitude is 50 mV,potential increment is 3 mV,pulse width of 0.05 s,pulse interval is0.3s;At the optimized conditions,the calibration curves for the determination of the phytosterol demonstrated an excellent linear response in the range from 160800?g/mL?r=0.998?,with detection limits 8.8?g/mL?S/N=3?,the proposed method was successfully applied to determination of phytosterol in corn germ oil deodorizer distillate,the recovery rate between 97.0101.3%.The functional nature of phytosterol was investigated,the results confirmed that corn germ phytosterol has the best antioxidant effect to delay lipid oxidation,when the dosage of the phytosterol in peanut oil was 0.08%;but also with TBHQ and VE has a synergistic antioxidant effect,and the synergistic antioxidant effect with TBHQ was the best.The clearance of DPPH·by phytosterol was not obvious;the maximum clearance of·OH was67.68%with the phytosterol concentration of 0.2 mg/mL;with the phytosterol concentration of 3 mg/mL,the maximum clearance of nitrite was 72.2%.In addition,the antibacterial effect of phytosterol was sdudied,the results show that almost no antibacterial effect on Staphylococcus aureus and Escherichia coli,Aspergillus and Mucor on significantly inhibition.
