Research Of The Preparation Technology And Quality Control For Olanzapine Tablets

Schizophrenia is a chronic and disabling illness with high frequency,which is difficult to treat completely Traditional antipsychotics generally block dopamine(DA)D2 receptors,which are the antagonist of D2 receptors.Compared with conventional antipsychotics,The second generation of antipsychotics can block not only the dopamine D2 receptors,as well as serotonin(5-HT)receptors.The second generation of antipsychotics have the characteristics of multi-target and more receptors,which are called atypical antipsychotics.Olanzapine is a derivative of thiophene benzo diazinon,which is developed by the American company of Lilly.The clinical trials show that olanzapine can improve clinical symptoms of schizophrenia with low side effect and better safety.This makes olanzapine become a best-selling antipsychotic in the world.This paper makes a study of preparation process and quality control on olanzapine tablets,which provides a basic research for the development of olanzapine tablets.Firstly the formulation and process of olanzapine tablets is carried on with the index of the preparation's fragility,disintegration time limit,angle of repose and bulk density.Based on the study above,the optimal formulation is determined.The result shows that the formulation is good,and the preparation has a nice disintegration,the olanzapine tablets are bright and clean.Secondly,in order to control the quality of the preparation of olanzapine tablets,the preparation's quality control is studied.HPLC methods,HPLC screening methods and the dissolution test are made for the study.Methodology and methods are validated.At last,the formulation,preparation and quality standard of olanzapine tablets are determined.
