| Selenium,as one of 14 kinds of trace elements necessary for humans and animals,can effectively clear the free radicals,enhance the body’s antioxidant capacity and immune function and reduce risk of cancer.Sodium selenite(SS)was widely used as supplement of selenium.Selenium-enriched yeast(SEY),produced by fermented Saccharomyces cerevisiae in an inorganic selenium-rich media,is a recognized source of organic food-form selenium.The kind of media not only provides selenium but also provides a certain amount of proteins,amino acids,vitamins and other nutrients.Se-methyl selenocysteine(SeMSC),derivatives of selenocysteine methylated,has advantages such as definite chemical structure,content stability,bioavailability and antitumor activity,compared with other organic selenium.Both VE and anthocyanin extracts of purple carrots(Ant)not only can lenify cardiovascular inflammation,but also assist the treatment of many diseases,such as atherosclerosis,inflammatory and cancer.Many studies have found that a variety of antioxidant factors in combination have better antioxidant effect than a single component.Synergistic effect can effectively conserve resources and greatly enhance the antioxidant capacity.Therefore,the antioxidant effects of individual and combined selenium,VE,anthocyanin extracts of purple carrots is worth further exploration.The antioxidant effects of individual and combined selenium,VE,anthocyanin extracts of purple carrots against the H2O2-induced oxidative stress in H9c2 cells,and against the D-Galactose induced oxidative damage in rats were investigated in this research.Meanwhile,the synergistic antioxidant effect of selenium,VE,anthocyanin extracts of purple carrots in vitro were studied.The main contents of the thesis are summarized as follows:1.PH-differential spectrophotometry was used to assess the anthocyanin content(10.42%)of purple carrots.Then the high-performance liquid triple quadrupole time of flight mass spectrometry(HPLC-QTOF-MS)was used to preliminary identify the structure of anthocyanins.2.The antioxidant effects of individual and combined selenium,VE,anthocyanin extracts of purple carrots were determined by DPPH radical scavenging activity,Fe3+ reducing power and ABTS radical scavenging activity.Equivalent surface analysis was used to evaluate the synergistic effect of individual and combined selenium,VE,anthocyanin extracts of purple carrots.The results indicated that SeMSC and SS possessed low in vitro antioxidant activity,while SEY possessed high antioxidant activity and exhibited synergistic effect with VE,anthocyanin extracts of purple carrots.3.H9c2 cells were randomly divided into 11 groups.After incubated for 24 h,H9c2 cells were treated by 400 uM H2O2 for 6 h.Cell viability was evaluated by MTT assay and the malonaldehyde(MDA)content,superoxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px)activities were measured.The results indicated that compared with model group,the individual and combined treatments of selenium,vitamin E,anthocyanin extracts could increase the viability of H9c2 cell significantly.Moreover,the MDA contents in cells were decreased and the activities of SOD,CAT,GSH-Px were increased.The changes of the combined groups were better than those of individual groups.4.Rats were randomly divided into 11 groups.All rats were treated with D-galactose solution(400 mg kg-1 BW per day)for 42 days to cause oxidative stress.At the termination of the experiment,the activities of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD),the contents of malondialdehyde(MDA),total antioxidant capacity(T-AOC),glutathione(GSH)and carbonyl protein(PCO)in the serum and tissues(liver,heart and kidney)were measured.It demonstrated that the individual SeMSC,SS,SeY,Ant and VE could significantly increase the activities of GSH-Px,SOD and the contents of T-AOC and GSH as well as decrease the MDA and PCO concentrations in the serum and tissues.For different forms of selenium,SeMSC and SeY showed greater antioxidant activity than that of SS at the same dose.Moreover,group(ANT+ Vit E)showed better antioxidant activity than the individual ANT and Vit E groups.Meanwhile,the antioxidant activities of groups(SeMSC +ANT + Vit E),(SS + ANT + Vit E)and(SeY + Ant + Vit E)were greater than those of groups(ANT + Vit E),(SeMSC),(SS),and(SeY)due to the synergistic effects of the individual antioxidants. |
