In the food industry,the pigment is an important food additives.Food coloring pigment include two categories,chemical synthesis and natural pigments.Because synthetic pigments have carcinogenic,teratogenic and other toxic,to some extent,limited its development and application,and therefore the higher the security of natural pigments gradually coming into view.Natural pigments can be divided by source of plant pigments,pigment animal,microbial pigments.Where animals and plants pigment quality differences large and small yield,high cost,and generally less stable,making application development is restricted.The microbial pigment as an emerging industry,get more and more attention of researchers.In particular,certain fungi will produce pigment(such as red niger)in the metabolic process,and fungi grow fast,easy cultivation can be carried out simply large-scale industrial production,so the fungal cytochrome becoming an important way to get the natural pigment.The laboratory,screened a strain of fungus Geomyces sp.WNF-15A from the Antarctic samples to the energy secretion purple hormone,in the Antarctic microbial screening process.Geomyces sp.WNF-15 A is a kind of Geomyces fungus and a fungal strain yielding purple pigment.In the pre-experimental study,we found that pretreatment of fermentation broth can obtain solid-color pigment powder which colour price reached 144.05.1.71 g of pigment powder can be obtained in 1L fermentation broth.Geomyces sp.the capacity producting pigment of WNF-15A has obvious advantages in similar microorganisms.In addition,it has a similar chemical structure with red yeast rice,and therefore with similar chemical stability.It has great development potential in the food additives industry.To achieve industrial production of Antarctic red pigment,the subject conducted more thorough research about Antarctic red pigment.Antarctic red pigment were isolated and purified by column chromatography and preparative HPLC techniques,combined with UV,IR,MS,HR-ESIMS,1H-NMR,13C-NMR,2DNMR(including HSQC and HMBC),etc.to analysis monomer compounds structural.and we found its mother nuclear structure is same the with monascorubrin.Therefore,to compare with red yeast rice,Antarctic red pigmen has further industrial development potential.Also,to overcome its shortcomings low temperature fermentation,we made UV mutagenesis to Geomyces sp.WNF-15A strain and optimization of culture conditions by appropriate surface method,in order to obtain high-temperature,high-yielding strains of pigment.I hope the Antarctic red pigment developed as a way to meet the food.pharmaceutical,cosmetics and other industries to adapt to multi-purpose pigments.The study found the Antarctic red pigment has big polarity,separation and purification difficult.We obtained monomer compounds of Antarctic red pigment successly by the anti-preparative separation.It is identified to determine that its chemical formula is C23H23N05,the same nucleus structure with monascorubrin nucleus structure.Research by the compare the stability of the red pigment and red yeast rice,found red pigment in Antarctica has good stability to UV.It is stable to the vast majority metal ions and food additives.And it has good stability range 6-10 at pH.Preliminary study by HPLC method found that Antarctic red pigment contain citrinin,security is better than red yeast rice.We found that Antarctic red pigment is nontoxic by the toxicology studies.And we got the high temperature pigment mutation producing strain YB-012 successfully,optimized by response surface methodology PH value obtained,the sugar concentration,the optimum value of the inoculum were 5,30%,7%.The results showed that the red pigment stability of the Antarctic,safer than red yeast rice,mutant strain of high temperature,high yield pigment,with the development of food,medicine,cosmetics industry colorants potential.
