In recent years medical applications of nanotechnology has attracted more and more attention from researchers,especially nano drug delivery system,because it's strategy,people spend a lot of focus on this area.Over the past decade,polymer drug carrier,including polymer-drug yoke close and polymer micelle was proved to be effective in the aspect of drug delivery,and several formulations have been applied to clinical trials,polymer micelle is now recognized as one of the most promising drug carrier.This experiment design was prepared with poly phosphine nitrile skeleton,polyethylene glycol monomethyl ether as the hydrophilic segment,dipeptide for hydrophobic chain segments and folic acid to targeted groups,through covalent bond cis monkshood anhydride and anti-cancer drug doxorubicin amphiphilic polymer drug carrier.Using nuclear magnetic resonance hydrogen spectrum(1H-NMR),infrared spectroscopy(FT-IR)and ultraviolet spectrum(UV)method for synthesis of a series of amphiphilic graft polyethylene phosphonic nitrile for structural characterization.Pyrene fluorescence probe method was used to study the amphiphilic polymer phosphonic nitrile self-assembly behavior in aqueous solution.According to the fluorescence intensity of pyrene fluorescence intensity ratio of I339/1333 figure and the relationship between the LogC figure,measured in the synthesis of the MPEG/GK(Boc),the lowest critical micelle concentration of Et-the PPP is 0.1033 g/L.Measured by uv spectrometry 1 g polymer containing folic acid 5.80 mg,1 g polymer contains 25.44 mg doxorubicin.Using dynamic light scattering(DLS)to study the amphiphilic graft poly phosphonic nitrile in aqueous solution,the particle size and distribution,preparation of final product particle size in 10-100 nm range.Studied the amphiphilic polymer phosphonic nitrile FA/DOX/MPEG/GKEt-PPP micelle in the pH value of 5.6,6.8 and 7.4 of the in vitro release of PBS buffer solution.Results show that the pH value of 5.6,6.8 and 7.4,the micellar within 48 h of the accumulative release percentage were 36.21%,28.24%,22.05%.
