Abalone Gonad Polysaccharide is a kind of sulphated polysaccharides extracted from abalone gonad.As reported the abalone polysaccharide have obvious biological activities.Now the structure-activity relationship of the polysaccharide is not completely clear,the structure information restricts the overall development level.In this study we used chemical degradation method on AGP-32,got the structure information of the main chain,and then reduced the difficulty of structure research.Finalize get the abalone gonad polysaccharide structure information.We use the double-enzyme hydrolysis method for abalone gonad polysaccharide(AGP),use Cellulose DEAE-52 and Sepharose CL-6B purified the polysaccharide,and then got the purified polysaccharide(AGP-32).After compared periodic acid oxidation method with partial acid hydrolysis method,we select the most optimal method for AGP-32 degradation.So we can get the main chain.Finally,by the method of periodic acid oxidation,Smith degradation,methylation and NMR analysis,we get the structure information.The molecular weight of AGP-32 is 86.3 kDa and the monosaccharide composition is Man,Rha,GlcUA,Glc,Gal,Xyl,Ara,Fuc,by the molar ratio of it is 5:1:2:1:10:2:1:8.Compared Periodic acid oxidation method with partial acid hydrolysis methods,we found the optimal degradation method of the main chain of polysaccharide AGP-32 was partial acid hydrolysis,the monosaccharide composition of the main chain are Gal,Glc,Man,and the molecular weight is 50 kDa.After periodate oxidation,Smith degradation we identified the core part of the composition of monosaccharide is Glc and Gal,connected by(1?3)glycosidic bond made up the main chain on a ratio of 3:1.After methylation,we can identify the result that it connected only by(1?3)glycoside bond.NMR analysis found that the Glc and Gal are both ? configuration.Finally,the main chain structure is:-[3)-?-D-Glc(1?3)-?-D Gal(1?3)-?-D-Glc(1?3)-?-D-Glc(1?]n-.
