Polysaccharides were extracted and purified from the abalone sexual gland and scallop viscus, and their immunomodulating and anticoagulating activity were investigated.Crude polysaccharide (CAGP) was extracted from abalone gland by alkali protease and pepsin treatment, the polysaccharide and protein content of CAGP were24.50%and45.15%, respectively. Crude polysaccharide (CSVP) was extracted from scallop viscus by papain hydrolysis, the polysaccharide and protein content of CSVP were45.08%and31.00%, respectively. The aboe two crude polysaccharides were further purified on a Sephadex G-100gel column, and thus abalone gland polysaccharide fraction AGP-1and scallop viscus polysaccharide fraction SVP-1were abtained. The polysaccharide, protein, sulphate radical and aldonic acid content of AGP-1were34.65%,33.60%,9.19%and18.02%. The contents of above four elements in SVP-1were48.73%,25.10%,5.14%and14.89%.In vitro results showed that,10-500μg/mL CAGP, AGP-1, CSVP and SVP-1could enhance lymphocyte proliferation and phagocytosis of peritoneal macrophages in a dose-dependent manner. AGP-1and SVP-1could also inhibite endogenous and extrinsic coagulation pathway, especially the former one, by prolonging ATPP and PT time.The in vivo immunomodulating acitivity of AGP-1was further investigated based on the results of in vitro experiments. Data showed that AGP-1could modulate the immune function of normal and immunosuppressive mice by enhancing thymus and spleen index, lymphocyte proliferation, phagocytosis of peritoneal macrophages, antibody production and the contents of CD3+、CD4+'CD8+T subgroups. Moreover, increase of the level of cytokines,such as IL-2、IL-4、IL-1β、TNF-α IFN-γ were also detected by ELISA assay, especially for IL-2、 IL-4and IFN-γ.The above data indicated that the polysaccharide from abalone gonad could strengthen the cellular and humoral immune function of normal and immunosuppressive mice. The scallop organ polysaccharide could enhance lymphocyte and peritoneal macrophages cell immune activity and anticoagulant activity in vitro, and its in vivo activity has yet to further research. Both abalone and scallop polysaccharide had similar anticoagulating activity against endogenous pathway. All the findings could be useful and helpful for the comprehensive application of the by products of abalone and scallop processing industry. |
