Eggplant is solanaceae genus annual herbaceous plants for perennial tropical,Has medicinal toxic heat stop bleeding,acetanilide detumescence,carbuncle sore,skin ulcer,hematochezia,treatment of frostbite,lower high cholesterol,high blowd pressure,prevention and cure stomach trouble,etc.Fruit edible part accounted for 93%.The biological activity of eggplant stick function has not been reported.Through various data analysis,the plant roots contain polysaccharide composition as the basis of a study.In this article,water extract-alcohol precipitation based on the traditional extraction method to study the extraction process of polysaccharide in eggplant stick,separation and purification technology and eggplant stick polysaccharides obtained by the physical and chemical properties and in vitro,in vivo antioxidant effect and immunoregulatory activity of study.In order to lay the ground for industrialization of eggplant stick polysaccharide.By the single factor and orthogonal experiment,determine the optimum conditions of polysaccharide in eggplant stick polysaccharide,using95%ethanol alcohol sinking operation.Finalized when the material liquid ratio of 1:8 0,extraction times for 1 time,extraction temperature is 70 ? and the extraction time was 2h in eggplant stick polysaccharide yield is the highest.At this point in eggplant stick polysaccharide yield of 2.87%,One of the factors affecting the yield of polysaccharide from primary and secondary order of material liquid than extraction temperature and extraction time.By the method of trichloroacetic acid,Sevag method,papain enzyme hydrolysis,enzymatic hydrolysis combined with trichloroacetic acid method,Enzymatic hydrolysis with Sevag method combined with the four methods of deproteinized experiments respectively,clearance rate and loss rate of polysaccharide to protein as evaluation indicators,comparing the several kinds of the separation of protein which is most appropriate.Results showed that different methods of take off the protein decreased with the increase of the number can increase protein removal rate,but the damage will be accompanied by a large number of polysaccharides.Combined with enzymatic,can be very good improve protein removal effect,at the same time reduce the loss rate of polysaccharides.Trichloroacetic acid method and the Sevag method respectively in papaya protein enzymatic combined deproteinization effect after all due to separate use,with enzymatic hydrolysis with trichloroacetic acid method protein clearance rate was 60.5%,the loss rate of polysaccharide is 57.8%,Compared with the other three methods,the effect is excellent.Take off the protein after polysaccharide in eggplant stick polysaccharide whit DEAE-D22 cellulose separation,ESP1 get three components,ESP1-2,ESP-3,including ESP 1-1 for the maximum elution peak.It by Sephadex G-100 gelcolumn chromatography for further the separation of test from ESP 1-1 was relatively single pure polysaccharide composition.The physical and chemical properties of eggplant stick polysaccharide ESP1-1 showed that the results indicate eggplant stick polysaccharide is pale yellow powder,-easy to dissolve in water,doesn't dissolve in organic reagent,phenol-sulfuric method of orange.Fehling precipitation reaction with red and green solution.It's show that contains free monosaccharide.A purple ring Molish response form,proof and aminoglycoside sugar.Iodine,potassium iodide response in blue,that does not contain starch,Ferric chloride reaction precipitation that does not contain phenolic substances,ultraviolet absorption peak at 280nm.Suggests that proteins were purified polysaccharide in eggplant stick polysaccharide still exist,this may be because theie are combined with polysaccharide of glycoprotein.By the thin-layer chromatography detection,the results show that mannose,flucose,fructose,monosaccharide composition,in order to further prove ESP1-1 for further analysis by high performance liquid chromatography(HPLC)method,the results showed that polysaccharide in eggplant stick polysaccharide braches except by mannose,glucose,fructose composition,also composed of xylose.In vitro antioxidant experiment showed that refined polysaccharides in eggplant stick polysaccharide branches ESP1-1 on superoxide anion and hydroxyl radicals have clearance ability,and the ability to clear as eggplant stick polysaccharide polysaccharide ESP 1-1 increase with the increase of the concentration,the mice experiment shows,ESP1-1 can increase SOD content in the body of MDAcontent,and compared with the model group has a significant antioxidant effect.Immune experiment results indicate that the polysaccharide in eggplant stick polysaccharide ESP1-1 could improve the ability of mice spleen lymphocyte proliferation test indicators,proved in eggplant stick polysaccharide can improve theimmune function of mice,reach and improve the effect of effect compared with model group significantly.
