This paper is mainly about the biotransformation of protopanaxatriol type ginsenoside from Panax notoginseng by enzyme.First,the total ginsenoside was extracted by methanol from root and fiber of notoginseng.Then the separation method of saponin monomer was researched.The reaction of transforming notoginsenoside Ri and ginsenoside Re to Rg1 was studied.At last,ginsenoside Re was transformed to ginsenoside Rg2,Rg4 and Rg6 Hydrolysate Rg2,Rg4 and Rg6 were separated by silica gel column and AB-8 macroporous adsorbent resin.338.50 g total ginsenoside was extracted by methanol from 4kg root of notoginseng,the yield of total notoginsenoside was 8.46%.66.50 g total ginsenoside was extracted by methanol from 4 kg fiber of notoginseng,the yield of total notoginsenoside was 1.66%.The content of total ginsenoside in root and fiber were similar based on HPLC,they were Rg1,Rb1,R1,Rd,Re.Results showed that R1.Rd,Re can't be separated by silica gel column,but Rg1,Rbl,R1,Rd,Re can't be separated by AB-8 and silica gel column.In the enzymatic reaction,the optimal microbe in optimal medium which can transform Re and R1 to Rg1 was screened.Then optimal condition for the enzymatic reaction was researched.Rg1 was transformed by enzyme from Re and R1,and this optimal condition for enzymatic reaction was:substrate concentration 2.0%,pH 6.0,at 37? for 48 h.Rg1 was transformed by enzyme from PPT,and this optimal condition for enzymatic reaction in the agitated reactor was:rotate speed was 90 r/min,concentration 2.0%,pH 6.0,at 37?for 48 h.Rg1,was separated by AB-8 macroporous adsorbent resin.At last,ginsenoside Rg2,Rg4 and Rg6 was obtained from ginsenoside Re,and its hydrolysate was separated by silica gel column and AB-8.Only a little of Rg4 and Rg6 can't be separated by silica gel column,but a lot of Rg2 can be separated by AB-8 with 42%ethanol.
