In recent years,research focus on the research of soybean peptide and soy oligosaccharide at home and abroad,soybean peptide yield is not high and product bitterness issues such as restricted the development of soybean peptide market.In this paper,edible soybean as raw material,choose alkaline protease,neutral protease,trypsin as the tool enzym,preparation of soybean peptide.Suitable enzymes species,ratio,pH,temperature,time are selected by the test,to achieve the purpose of appropriate increase in the yield of hydrolysis and reduce bitterness.Edible soybean as raw material,alkaline extraction method for extracting the soy protein isolate best production process is:solid-liquid ratio 1:12,pH9.0,temperature 55?,time 75 min,extraction rate of 89.45%,under this condition,the extraction of soybean oligosaccharide content of 10.11%.Through single factor and orthogonal test,optimization of alkaline protease,neutral protease,trypsin digestion process,compared to hydrolysis yield and bittemess.The best conditions for the neutral protease is pH8.0,Temperature 45 ?,enzyme dosage 7500u/g,time 4.5h,hydrolysis yield of 81.26%,bittemess is 7.The best conditions for alkaline protease enzyme is pH10.0,the amount of enzyme 10000u/g,temperature 55 ?,time 4.5h,hydrolysis yield of 91.24%,bittemess is 9.Trypsin optimum conditions for pH8.0,the amount of enzyme 7500u/g,temperature 50?,time 4.5h.Based on the best hydrolysis conditions,hydrolysis yield of 79.04%,the bitterness is 7.Compounded three protease,optimizing enzymatic hydrolysis process by orthogonal experiment.The optimal hydrolysis conditions of Alkaline protease and neutral protease is:alkaline Protease:neutral protease enzyme ratio 1:1,pH9.5,temperature 50 ?,time 4.5h.Based on the best hydrolysis conditions,hydrolysis yield of 85.06%,bittemess is 5.The optimal hydrolysis conditions of neutral protease and trypsin is:neutral protease:trypsin enzyme ratio 1:1,pH8.5,temperature 50?,time 4.5h.Based on the best hydrolysis conditions,hydrolysis yield of 81.09%,bittemess is 4.The optimal hydrolysis conditions of alkaline protease and trypsin is:alkaline protease:trypsin enzyme ratio 1:1,pH8.5,temperature 50?,time 4.5h.Based on the best hydrolysis conditions,hydrolysis yield of 83.33%,bittemess is 5.The optimal hydrolysis conditions of alkaline protease,neutral protease and trypsin is:alkaline protease:neutral protease:trypsin enzyme ratio 1:2:3,pH8.5,temperature 50?,time 4.5h.Based on the best hydrolysis conditions,hydrolysis yield of 86.76%,the bitterness is 2,soy oligopeptides content is 93.49%.Three enzyme complex can not only to improve the yield of hydrolysis,but also greatly reduces the value of hydrolyzate bitterness.Therefore,a three-enzyme complex enzymatic method is more suitable for the production of high yield,low bitterness soybean peptide on the industry.
