Abalone has been loved by customers for its rich amino acid and low cholesterol and fat.It is well known as "the champion of seafood",and it has a good reputation in the international market.In recent years,with the increase of abalone production in our country,lots of abalone viscera have been abandoned,and only a small number of viscera are processed into fodder.Studies have shown that the activity of polysaccharide extracted from abalone viscera have many functions such as antioxidant,lower fat and blood sugar,and so on.Our team has extracted crude polysaccharide from the abalone viscera at earlier stage,and verified its physiological activity,but the mechanism of abalone polysaccharide is uncertain.In this study,the abalone viscera polysaccharide was degraded by solid phase acid,then isolated and purified oligosaccharide for further analysis of its nature,structure and antioxidant activity.The main tasks are as follows:1.Use low speed centrifugation instead of frame filter to improve the extraction process of polysaccharides,reduce the loss of raw materials during the operation.Use Sevage method to remove the protein,it can remove 80%protein off after handling six times,achieve better results.After bleaching treatment with activated clay,polysaccharide significantly become lighter color,then go through the resin filter,polysaccharides almost colorless,indicating resin also has bleaching effect.732 bonded with 717 anion resin activated clay treatment can remove the heavy metal well.2.1n order to study the relationship between the condition of hydrolysis and the yield of hydrolysis rate,three factors were chosed to optimize the yield of hydrolysis rate using the method of Response Surface,they were concentration of hrdrolysis temperature,hrdrolysis time and the quantity of resin.Regression equation,Y=80.69+0.61X+1.91X2+2.75X3-0.66X1X2+0.027X1X3-0.81X2X3-3.15X12-3.31X22-3.30X32,was fitted by multiple linear regression.From the regression equation,among the three factors,the effect on the yield of hydrolysis rate was concentration of hydrolysis temperature>hydrolysis time>the quantity of resin.The validating test was carried under the three factors level,and the result showed that there was no significant difference between the test-value and the forecasting-value(P>0.05).3.Oligosaccharide fractions were isolated by gel permeation chromatography,and analyzed its physical and chemical properties,the result showed it contains 4.9%of SO42",8.9%of glucuronide,molecular weight was 2300.IR analysis demonstrated that the backbone of AVO was a-type glycosidic linkages,and it contained pyranose.4.Different antioxidant models have been employed to acquire the antioxidant activity of abalone polysaccharides and abalone oligosaccharides.The result showed that oligosaccharides has a higher antioxidant activity compared with abalone polysaccharides in different models.But they are both weaker than Vc.
