The Preparation Of Microparticles Embed Nisin And The Antimicrobial Effect Of Embed Nisin

Nisin is a natural antimicrobial peptide produced by strains of Lactococcus lacts subsp.Lactis and it has received particular attention.Although Nisin has been used as the natural antiseptic substance,there is still a big weakness that the vulnerable degradation in fluids lead to shorten its shelf-life.In this study,in order to improve Nisin's stability and control its degradatio,the embed of Nisin on ?-CD and antibacterial property of embed Nisin were investigated.Using ?-CD to prepare Nisin-loaded clathrate compounds,the optimum formula was found according the drug loading capacity.Then prepared the gelatin-arabic gum according the optimum conditions.In this study,antibacterial property of embed Nisin was investigated.According to the drug loading,we found the optimum conditions were as fellows:the ratio of Nisin and excipients is 4:6,added 50mL deionized water and stirred for 1h at 60?,dried in vacuo at 30 ? for 48h.The compounds prepared under this optimum condition have embed 63.60%Nisin.The embed Nisin was prepared under the optimum condition,and then the antibacterial activity were investigated.The results showed that embed Nisin achieved better and longer bactericidal activity than free Nisin.Under the same condition,embed Nisin showed bactericidal time longer than free Nisin in bacterium solution at 30?.Embed Nisin and free Nisin sterilized by 121? for 5 min and 10 min were investigated based on their bacteriostatic effect with inhibition zone method.The results indicated that embed Nisin showed better bacteriostatic effect than free Nisin.Form bacteriostatic effect of sustained release of embed Nisin in the buffer solution,embed Nisin would increase sustained release time.The Nisin microcapsule prepared in gelatin and arabic gum has showed excellent bacteriostatic effect and sustained release bacteriostatic effect,too.The two kinds of embed Nisin,solved the problems that the vulnerable degradation in fluids lead to shorten Nisin's shelf-life,and the result also showed excellent antibacterial effect and sustained-release bacteriostatic effect.During the processes of preparing embed Nisin,we haven't using organic solvent,while lengthen the bacteriostatic time and solve the problem of Nisin's weak stability.Thd embed Nisin has some certain actual application value.
