The sea cucumber have high nutritional and medicinal value as medicine-food marine products.As a part of sea cucumber,the viscera of sea cucumber have higher value.However,in China,they were not been used or full used but just disposed as the scraps during the process of sea cucumber,which caused great waste of resources and environmental pollution.The viscera of sea cucumber contains a variety of active substances such as the polysaccharide,the polypeptide and the saponins,and so on.The bio-active substances have various physiological functions,including anti-oxidant,anti-fatigue,immunity enhancement,and so on.Utilizing viscera of sea cucumber as raw material in this paper,the best exogenous protease was selected to hydrolyze the viscera of sea cucumber,the biochemical indicators of enzymolysis liquid during the process of digestion was also studied at the same time to certain the optimal conditions of enzymatic hydrolysis.The dosage by gavage in mice was confirmed according to the protein concentration by measuring the nutrients in hydrolysis products.The oxidation activity and anti-fatigue activity of the hydrolysis product of the sea cucumber viscera can be determined through animal experiments.All of these could lay theoretical basis for the deep processing of the sea cucumber viscera.The main research contents and conclusions were summarized as follows:1.The optimal enzymolysis technology of sea cucumber viscera was ascertained through the single factor experiments and response surface method determined:the best neutral protease was selected in this experiment as hydrolysis enzyme,The best hydrolysis process:the adding amount of 2000 U/g,liquid-solid ratio 12:1 and temperature 50?,pH7.Hydrolyzate over 10000Da membrane,ultrafiltration through concentrated and dried to get hydrolyzate of sea cucumber viscera,where protein or polypeptide content of 73.02%,5.96%total sugar,saponin 1.39%,13.09%ash content,water content 7.2%;2.Hydrolyzate of sea cucumber viscera showed obvious antioxidant activity in vitro,the IC500 of the·OH radical and DPPH·radical were 1.26 mg/mL,5.33 mg/mL.Anti-oxidant in vivo showed that oyster peptide to increase antioxidant capacity had very significant effect:the low?0.2 g/kg·BW??middle?0.4 g/kg·BW?and high group?0.8 g/kg·BW?had no obvious effect on mice weight,the enzymatic hydrolysis products of sea cucumber viscera could not only significantly reduce the MDA content in serum and liver?P<0.05?,but also increase the T-SOD and GSH-PX activity in serum and liver?P<0.05?.They had better antioxidant activity.3.The results of Anti-fatigue activity by animal experiment proved that the enzymatic hydrolysis product of sea cucumber visceral had no obvious effect on mice weight,they could significantly reduce serum lactic acid and urea nitrogen content of trained mice,the difference had statistically significant?P<0.01?;liver glycogen of three dose groups were 7.19?8.71?6.98mg/g,increased by 30.68%?58.41%and 26.92%than normal group?p<0.01?.This showed that the hydrolyzate of sea cucumber viscera had better anti-fatigue activity.
