Gold nanorods(GNRs)have good and unique optical properties,and it can control the ratio of length to diameter,to produce different anisotropy and unique spectral features,so it is widely used in the antibody identification,nano-materials assembly,cancer cell imaging and DNA analysis.With the development of science and technology,study on the signal amplification and photothermal diagnosis and treatment of GHRs is still deepening,in order to establish a mature and safe diagnosis and treatment strategies and techniques.In this paper,a nano-probe based on gold nanorods and aptamers was constructed to study its application in biosensor and tumor photothermal therapy.It was divided into the following four chapters:The first chapter summarizes the history and properties of gold nanorods,the introduction and application of aptamers,the preparation,assembly and application of gold nanorods and aptamers in the field of biology.On this basis,this paper puts forward the basis of this paper,research purpose and research content.In the second chapter,Apt29/thrombin/Apt15 sandwich structural sensor was constructed based on thrombin-based apt29 and Apt15 for the high sensitivity test of thrombin.The change of extinction value and the calibration curve of thrombin concentration were realized by metal waveguide capillary photometer:?EMWC=2.21×10-4c(nmol/L).The linear range was 0.068-1.09 nmol/L(R = 0.971)and the detection limit was 0.02 nmol/L.Compared with the measurement results of ordinary spectrophotometer(1 cm cuvette),the detection limit is improved by 68 times.The application of metal waveguide capillary photometer in the field of analytical chemistry was extended.In the third chapter,GNRs-AptRomas probes were prepared based on the aptamers of human lymphoma Romas(AptRomas)for quantitative spectrophotometric detection of Romas cells in vitro.The absorbance of GNRs was inhibited by the combination of AptRomas and GNRs.After addition of Romas cells,AptRomas and the target Romas cells were combined.The gold nanorods were released,and return to the initial absorbance values.The linear equation of Romas cells is:E = 0.01351 gC(cell/mL)+0.0283,R=0.967,the detection limit of up to 5 cells/mL.In the fourth chapter,GNRs-Apt A549 probe was prepared based on human lung adenocarcinoma A549 cell aptamer(Apt A549)to explore the effect of gold nanorod on cell proliferation of human lung cancer cell A549 in dark and light.The experimental results showed that It can overcome the cytotoxicity or damage of the cell membrane caused by the introduction of CTAB in the traditional synthesis process,at the same time,its thermal conversion ability has not decreased.The concentration of GNRs has some influence on the photothermal treatment.The targeting effect of Apt A549 needs further study.
