Walnut(Juglans regia L.)is a comprehensive development and utilization of high-value woody and oil crops.Walnut shells are byproducts in walnut processing,generally as waste disposal or incineration,resulting in waste of resources.The main components of walnut shell is lignin,hemicellulose and cellulose,the main component of wood vinegar of walnut shells is acids,phenols,flavonoids,aldehydes,and some heterocyclic compounds.Searching for natural antioxidants and weight loss drugs is imperative.The antioxidant and anti-obesity properties of extracts from walnut shell were analyzed.The main contents and results were as follows:1.The extraction yield,total flavonoids content and total phenols content of various solvents used for the extraction of walnut shell showed significantly different(p<0.05).The N-butanol extract have the highest extraction yield and the extraction yield is 4.54%.The ethyl acetate extract showed the highest amount of phenols and flavonoids,with 200.40 mg gallic acid equal/g extract and 80.40 mg rutin equal/g extract.2.Ethyl acetate extracts have the strongest DPPH radical scavenging capacity and chloroform extract have the weakest DPPH radical scavenging ability.Methanol extract have the strongest hydroxyl radical scavenging ability and the ethanol extract have the weakest hydroxyl radical scavenging capacity.Chloroform extract have the strongest superoxide anion radical scavenging capacity and N-butanol extract have the weakest superoxide anion radical scavenging ability.The order of total antioxidant capacity from strong to weak is methanol extract>ethyl acetate extract>chloroform extract>butanol extract>ethanol extract>water extract.The order of reducing power from strong to weak is methanol extract>chloroform extract>water extract>ethanol extract>ethyl acetate extract>n-butanol extract.3.The order of inhibitory sequences is methanol extract>ethyl acetate extract>ethanol extract>aqueous extract>butanol extract>chloroform extract,respectively under the concentration of 500 ?g/mL.The IC50 values of water extract,methanol extract,ethanol extract,chloroform extract,n-butanol extract and ethyl acetate extract are 371.35?g/mL,308.91?g/mL,407.57?g/mL,196.50?g/mL,247.79?g/mL and 254.27?g/mL.The smaller of IC50 value indicated the stronger inhibition.With a standard of ICso,the descending order of the inhibition of walnut shell extract on pancreatic lipase is chloroform extract>butanol extract>ethyl acetate extract>methanol extract>water extract>ethanol extract.The larger of 1/Ki value indicated the stronger inhibition effect on the enzyme activity.With a standard of 1/Ki,the descending order of the inhibition of walnut shell extract on pancreatic lipase is consistent with the results with a standard of IC50.4.All the extracts are less-toxic to 3T3-L1 cells.The chloroform extracts and N-butanol extracts have inhibitory effect on triglyceride accumulation of 3T3-L1 cells.The extract of walnut shell can develop into potential drugs to treat obesity.
