As an important post-translational modification,protein glycosylation plays an important role in microbial infection,cell differentiation,tumor metastasis,cell carcinogenesis and other life activities.As a new technology of glycoprotein detection,agglutinin chip has attracted extensive attention due to its high throughput,convenience and rapidness.For the production of lectin microarray samples by fluorescence,with evanescent wave fluorescence signal detection of active detection technology.Nowadays,most of the dyes used for fluorescent labeling are cyanine dyes,but their synthetic routes are complex,cost is high,water solubility is poor,and stability is decreasing with the increase of methylene chain.Therefore,must exclude by rigid cyanine dye labeled glycoproteins by fluorescent dye impression,the synthesis of modified model,to remedy the defects,is of great significance.In order to obtain fluorescent dyes with marked protein capability,we designed and synthesized two kinds of fluorescent dyes containing six kinds of oxygen heterocyclic structures,and studied their properties and applications.It mainly includes the following two parts.1.Two new fluorescent dyes YD-1 and YD-2,and four rigid fluorescent dyes YN-1,YN-2,YH-1 and YH-2,which are mainly composed of heterocyclic structure,were synthesized.And new fluorescent dye YB modified by Rhodamine B was synthesized.The HRMS(ESI),1H NMR,13 C NMR and IR of the seven new compounds were characterized by detection;The spectroscopic properties of the other six kinds of dyes except YB have been detected.It is proved that all the six dyes have good light resistance;wherein,YD series of fluorescence emission wavelength near 600 nm,Stokes shift at about 80 nm the emission wavelength of fluorescence;YN and YH series of more bias in the near infrared region(above 600nm),Stokes displacement in 60-80 nm,that can reduce the interference of fluorescence imaging in biological luminescence caused by autologous effectively;according to the effect of p H on the fluorescence intensity of the dye has stable fluorescent properties in vivo in the range of p H.2.Six kinds of novel fluorescent dyes(YD,YN and YH three series)used in glycoprotein markers and achieve good results,indicating that these dyes can covalently bind with protein to glycoproteins with fluorescence signal analysis on the types of sugar chain protein after making the biological application of lectin microarray is of practical value.
