In this paper,a cotton thread immunochromatographic assay device based different carbon nanomaterials probe was constructed for rapid,effective,sensitive and visual detection of lung cancer biomarkers.The main works are summarized as follows:(1)Researches on cotton thread immunochromatographic device based on carbon nanotube decorated gold nanoparticles reporter probeIn this work,an immunochromatographic assay device based carbon nanotube decorated gold nanoparticles(CNT/GNPs)reporter probe was developed for rapid,sensitive and visual detection of a lung cancer related biomarker—Squamous Cell Carcinoma Antigen(SCCA).CNT/GNPs nanocomposite probe was applied to construct an immunochromatographic assay device on cotton thread.CNT/GNPs combined the advantages of carbon nanotube(CNT)and gold nanoparticles(GNPs),and exhibited good biological compatibility and higher stability.Compared with two reported immunochromatographic assay methods which used individual carbon nanotubes as reporter probe,this biosensor was 10 and 2000 folds more sensitive than previous reported CNT based lateral flow assay respectively.(2)Multianalyte Immunochromatographic assay on cotton thread for simultaneous detection of Squamous Cell Carcinoma Antigen and Carcinoembryonic AntigenOn the basis of the experiment(1),we designed an immunochromatographic assay based CNT/GNPs nanocomposite probe for simultaneous detection of Squamous Cell Carcinoma Antigen(SCCA)and Carcinoembryonic Antigen(CEA)in a rapid and visual manner.Using this approach,the color intensity difference is proportional to the analyte antigen concentration,and a liner ranges over 50-5000 ng/mL analyte were obtained.The biosensor was detectable at a limitation of 12.0 ng/mL SCCA and 15.6 ng/mL CEA based on S/N?3.(3)Study on the cotton thread immunochromatography device based on carbon nanospheres probe.In this paper,an immunochromatographic assay device based carbon nanospheres(CNSs)was developed for visual detection of SCCA.CNSs were a new member in carbon nanomaterials.The sensitivity of this device was improved clearly because of its excellent properties.The detection limits were 0.65 ng/mLbased on S/N?3.
