| Objective Based on the pharmacological effects of Jerusalem artichoke,licorice,and Gynostemma pentaphyllum polysaccharides,the hypoglycemic and lipid-lowering health tablets were developed The extraction conditions of polysaccharides from Jerusalem artichoke,licorice and Gynostemma were optimized,and the polysaccharide content was determined.Taking the extract polysaccharide as the main drug,screening the amount of excipients,determining the best prescription for the tablet,and conducting quality research to further establish the quality standard of the tablet.Methods Single factor experiments and orthogonal test design methods were used to investigate the effects of ratio of feed to liquid,temperature,time,number of times and ratio of alcohol precipitation on the yield of crude polysaccharides of Jerusalem artichoke,licorice and Gynostemma pentaphyllum.Ultraviolet spectrophotometry was used to determine polysaccharide content in crude polysaccharides.Tablets are prepared by wet granulation tableting,and the amount of fillers,disintegrants,and binders in tablet formulations are screened to determine the best prescription.The tablets were examined in accordance with the requirements of the tablets in the 2015 edition of the Pharmacopoeia and qualitatively identified by thin-layer chromatography.The content of polysaccharides in the tablets was determined by phenol-sulfuric acid method.Results The optimum extraction process of Jerusalem artichoke polysaccharide was 1:30 ratio of material to liquid,extraction temperature was 90℃,extraction time was 40 min,extraction frequency was 2 times,alcohol precipitation ratio was 1:4,crude polysaccharide yield was 49.4%,and glycyrrhiza polysaccharide was the most The optimum extraction process was material-liquid ratio 1:20,extraction temperature 90℃S extraction time 60 min,extraction times 2,alcohol-to-slover ratio 1:5,crude polysaccharide yield 11.8%,Gynostemma pentaphyllum polysaccharide extraction process as material liquid Ratio 1:20,extraction temperature 90℃,extraction time 60 min,extraction times 2,alcohol precipitation ratio 1:3.The crude polysaccharide yield was 9.7%.The contents of polysaccharides(polysaccharide/crude polysaccharide)of Jerusalem artichoke,licorice,and gynostemma were 0.90 g/g,0.49 g/g,and 0.15 g/g,respectively.Tablet Recipe,Jerusalem Artichoke Crude Polysaccharide:Licorice Crude Polysaccharide:Gypenoside Crude Polysaccharide:Microcrystalline Cellulose:Carboxymethyl Starch Sodium = 50:15:10:20:5,Add 1.5%PVP(K30)80%Ethanol Solution qs,Wet The method of granulation,dry particles and then add appropriate amount of magnesium stearate(about 0.2%of the total),tableting.The mass analysis showed that the average tablet weight was 0.403 g,the tablet weight was acceptable,the disintegration time was within 15 min,and the average hardness was 45.6 N.The total polysaccharide content in the tablets was 0.215 g/tablet.The limit for the formulation of polysaccharides in tablets is "This product contains glucose per glucide,not less than 0.2 g."Conclusion The extraction methods of Jerusalem artichoke,licorice root and Gynostemma pentaphyllum polysaccharide are stable and feasible.According to the best prescription of polysaccharide tablets determined by experiment,the polysaccharide surface with smooth and round surface can be obtained.Through the qualitative identification of tablets,preparation inspection,content determination and other tests,the quality standards of polysaccharide tablets were initially established. |
