Chinese white olive(Canarium album Raeusch.)and plum(Armeniaca mume Sieb.)these two food and medicine fruits were used as the main materialscombined with the dried tangerine peel,japonica rice and tea to make the compound oral liquid in this research.The extraction condition,composition analysis by thin layer chromatography(TLC)and pharmacodynamics of antitussive and expectorant effect of compound oral liquid of Chinese white olive and plum were studied.It could provide the theoretical basis for the further development of the compound oral liquid above.The results were as follows:1.Water extraction and alcohol precipitation were used to optimize the extraction processes of compound oral liquid of Chinese white olive and plum.Extraction time,ratio of material to liquid,times of extraction were selected as investigation factors by using the rutin as standard sample.Orthogonal test was used to optimize the water extraction and alcohol precipitation process conditions.The contents of total flavonoids and the extraction rate were used as the evaluation indices.The results showed that the optimum water extraction process was heating extraction 3 times,extraction time 90 min,the ratio of material to liquid 1:10.The optimum conditions of alcohol precipitation were the concentration of liquid was 0.35 g·mL-1,the concentration of alcohol was 60%and the time of alcohol precipitation was 12 h.The extraction of total flavonoids by water has the advantages of short time,high extraction efficiency,safety and non-toxicity,simple operation and no need for high temperature.It was suitable for industrial production and large-scale production and application.In alcohol precipitation experiments,the alcohol concentration is relatively small and the time of alcohol precipitation is shorter.It was suitable for high efficiency production in factory under considering the factors such as the cost and the working hours.2.The constituent of the compound oral liquid consisted of the Chinese white olive(Canarium album Raeusch.),plum(Armeniaca mume Sieb.)and dried tangerine peel were analyzed by using thin layer chromatography(TLC).The results of TLC showed that the extraction samples of oral liquid had the same color spots in the corresponding position with each stand reference substance,which indicated that Qingmei compound oral liquid contained the above three kinds of fruit components.3.In this experiment,the cough model of mice was established by the coughing method of ammonia water,and the rat model was established by using the mouse tracheal phenol red excretion method.The antitussive and expectorant effects of compound oral liquid of Chinese white olive and plum in high,middle and low doses were observed.Studies show that oral liquids significantly reduce the number of coughs in mice within 2 minutes(p<0.05 or p<0.01),prolong the latent period of cough in mice(p<0.05),and promote the secretion of phenolic red(p<0.05).Therefore,Qingmei compound oral liquid based on this research had antitussive and expectorant effect.
