As an important research field of chemical biosensors,fluorescent probe have attracted extensive attention for its advantages of convenience,ultra sensitivity,noninvasion and spatiotemporal resolution.Based on these studies,in this paper,we constructed several BODIPY-based fluorescent probes and systematically introduced tiheir application in vitro and in vivo.There are plenty of reactive small molecules in organism,they are maintaining the organism working smoothly,and an abnormal level of these small molecules in living system always accompany with various disease.Thus,developing an effective method for monitoring these small molecules in living samples holds great potential for disease diagnosis,treatment and management Nitric oxide(NO)is an important signal molecule in living system,and nitroxyl(HNO)is a one-electron reduced and protonated derivative of NO,which plays a crucial role in many physiological and pathological processes in living organism.Nevertheless,HNO is an active molecule with poor stability in biologically relevant samples,which makes the detection of HNO challenging.In fields of living system,environmental monitoring and chemical manufacturing,the pH value is an important parameter,and has a wide impact For exanple,normal tissues and organisms have a relative stable pH values,while the pH value of cells microenvironment will be dramatically changed in disease tissues.Therefore,developing an effective method to accurate detection of pH valus is of great importance.At the same time,pH is also an important parameter in environmental monitoring.It is extremely common to cause damage to the ecological environment Thus,the research ofpH pH of great potentials for various research fields.Based on these facts,we developed several BODIPY-based fluorescent probes for accurate detection of small molecules and pH valus.The following is a brief introduction of the related content:1)We developed a novel fluorescent probe P-BODIPY-N based-on HO-BODIPY-Cl.After specially fine-tuned the electronic effects of the probe,P-BODIPY-N realized the specifically recognition of HNO in ratiometric model.We chose diphenylphosphinobenzoyl as the trigger moiety,which can impressively increase the anti-interference from biological reductants.However,both diphenylphosphinobenzoyl unit and organic small molecule have poor water-solubility,which extremely limited the application of the probe.Therefore,we encapsulated P-BODIPY-N into an amphphilic copolymer(mPEG-DSPE)and prepared a novel BODIPY-based micellar nanoprobe:NP-BODIPY-N.The new probe exhibits ultrasensitivity and good biocompatibility.It shows favorable cellular uptaken and was successfully used to detect intracellular HNO released by Angeli's salt in living cells and zebrafish larvae.2)We developed a novel fluorescent probe for pH measurement.The probe realized its pH(pH=9.06?11.76)detection via the fluorescence changes based-on the structure transfer between HO-BODIPY-N and-O-BODIPY-N.We hope the accurate detection of pH valus will provide some reference for the related research.
