| Paper-spray Mass Spectrometry(PS-MS)has been reported 9 years ago,which is a kind of Ambient Ionization Mass Spectrometry(AMS).PS-MS is a simple,rapid,solvent-efficient and high-throughput analytical method for analyzing complex samples.In addition,PS-MS method also has the advantages of simple sample preparation,sample loading and analyte extraction.Toosendanin fructus,come from the dry ripe fruit of Meliaceae deciduous tree chinensis,is a traditional Chinese herb listed in the Chinese Pharmacopoeia.In this article,three works were carried out and the results are showed as follows:1.The HPLC fingerprints of Toosendanin fructus were obtained by HPLC,and the seven chemical constituents of Toosendanin fructus were confirmed by LC-MS/MS,and quantitative analysis was carried out to make the chemical information of Toosendanin fructus clearly.Finally,in combination with ChempatternTM multivariate analyses,25 batches of Toosendanin fructus and 10 batches of Melia azedarach(an adulterant of Toosendanin fructus)were analysed,including similarity analysis and principal component analysis(PCA)the HPLC fingerprints.The similarity between Toosendanin fructus and common pattern was 0.88-0.98,which indicated that the fingerprints of Toosendanin fructus from different batches were relatively stable.The similarity of the nine samples of Melia azedarach with the common pattern was very low,which is between 0.03-0.55.They are clustered in two relatively discrete groups.The PC1 and PC2 scores were high(91.33%and 23.7%,respectively),which proved that the identification of species could be achieved by HPLC.2.A PS-MS method was developed to obtain MS profiles of Toosendanin fructus in positive and negative ion modes.By optimizing the paper matrix modified by different salt solutions,cone voltage and spray voltage,the mass spectrum profile can be obtained.In combination with similarity analysis and PCA,the PS-MS profiles of 25 batches of Toosendanin fructus are different from 10 batches of Melia azedarach.In the positive and negative mode,the similarity was better than that of HPLC.The score of PC1 and PC2 were slightly smaller than that of HPLC,but the difference was still very obvious.Therefore,similarity and PCA of PS-MS data are suitable for the categorization of Toosendanin fructus and Melia azedarach,and PS-MS method is far faster than HPLC method.3.By optimizing the excipients,temperature and other conditions of the processed method and combining with HPLC analysis,the difference between the 20%vinegar at 130℃ is the most obvious.The GC-MS fingerprint shows that the volatile oil constituents of Toosendanin fructus changed before and after processed.Finally,the differences of PS-MS fingerprints before and after the processed of Toosendanin fructus were analyzed.In combination with the similarity analysis and PCA of ChempatternTM,the rapid identification of 25 batches of Toosendanin fructus and 10 batches of processed drugs can be achieved,and the difference is very obvious.The results showed that the chemical composition of Toosendanin fructus changed after vinegar processed. |
