Monalosialic acid four hexose Ganglioside(GM1),as the main raw material and intermediate,is mainly used in the treatment of nervous system diseases such as the loss of central nervous system and Alzheimer's disease,and it has a growing market demand.The existing preparation process,through the extraction of total gangliosides from the pig brain tissue,and then the process of saponification,macroporous resin adsorption,hydrolysis conversion,chromatography purification and other steps of purification and purification,finally obtained pure products.It has a long process,long time consuming,large amount of organic solvent and unfriendly people to the environment.As a result,it is necessary to study a more simple,economically feasible,process safe and environmentally friendly preparation method,and then evaluate the safety of the preparation process,so as to provide a theoretical basis and reliable data for the application of the preparation process to the actual production,and reduce the damage to personnel and the impact on the ecological environment by the preparation of the process..The contents and results of the study are as follows:(1)Taking the maximum amount of total ganglioside as the target,the extraction conditions were obtained by single factor test and Box-Behnken response surface optimization.The results showed that the best yield of total ganglioside in the extract was when the solvent of 20% water content,the amount of solvent 10 times the ratio of 10 times and the temperature of 26(2)The optimized extraction of total ganglioside was treated by anion exchange chromatography.The results showed that the anion exchange resin had better adsorption and elution for total gangliosides when the amount of D201 resin,the flow velocity of 2BV/h,the eluent of 50 mM in the salt concentration and the elution velocity of 3BV/h in the 1/40 BV times of the pig brain extract.Effect.(3)By comparing the improved process with the existing process flow,amount and time,the results show that the amount of the 3 main solvents is more than 20%,the preparation cycle is shortened by 28.28%,and the recovery rate of the product is increased by 9.16%.At the same time,all the indexes of the GM1 products produced by the improved process meet the requirements of the national standard for the quality of GM1 APIs.(4)Using the simulation analysis of the aftermath of the pool fire accident and the quantitative calculation of the poisoning accident,the risk assessment of the process of GM1 preparation is carried out.The results show that it is effective to rationally plan the area of the dike in the tank area,to strictly control the exposure of the organic solvent in the environment,to improve the ventilation strip in the working environment,and to strengthen the protection of the personnel.Reduce the safety risks of the process to the personnel and the environment.
