| With the continuous developing of the food industry,people pay more attention on food safety.As reported salmonella can be survival in all kinds of foods.As one of the most common find food-borne pathogens,it is very important to detect salmonella in food safety.Traditional methods of detection salmonella has many disadvantages,such as cumbersome plate culture,complex medium preparation,long incubation time and so on,but it was difficult to satisfy the needs of food industry.It is very important to develop a new method to solve these problems.Fast test sheet was a better way to detection the salmonella with the low cost and widely using in factory.The culture medium of salmonella plays a major role in this method.In this work,culture medium of salmonella rapid test medium was investigated.First determination of the best content of each component,single factor experiment and orthogonal experiment were carried out to optimize conditions.After carefully experiments,the best conditions were as follow: tryptone 10 g/L,4 g/L yeast powder,sodium pyruvate 100 mg/L,150 mg/L niacinamide,bile salts of 3g/L,12 g/L sodium citrate,bright green 300 mg/L.Then the verify the validation of the part including sensitivity,specificity,medium the best training time authentication and identity authentication with national standard method of testing results,the results show that when the colony count of 0-9 can still be accurate count by the medium with high sensitivity,it was shown a strong specificity in interference bacteria.The optimal incubation time was 24 h.Compared with national standard method(kappa > 0.75),there was not any significant deference.According to the experimental results,it can speed up the detection of salmonella,improve the detection efficiency,and it would have high practical value in the detection of salmonella in the area of food safety. |
