Myopia has become a worldwide public health problem.The current situation of the myopia population in China is also not optimistic.It has reached nearly 400 million,of which young people account for the vast majority.Atropine can effectively control the progression of myopia,with slight adverse reactions and higher patient acceptance.However,traditional atropine ophthalmic preparations include eye drops,eye ointment and the like,which have the disadvantages of low bioavailability,strong irritation,and the like.In this topic,atropine sulfate is made into an inverse thermosensitive gel,which overcomes the traditional ophthalmic preparations.The shortcomings of the medicine method can also realize the automatic perception of temperature changes in the eye.The basic prescription chosen for this topic: methyl cellulose,polyethylene glycol,and sodium citrate.Determination of the best prescription was based on indicators such as phase transition temperature,gelation time,viscosity,and centrifugal stability.The release behavior of the preparation was studied by the non-membrane stripping method,and corneal permeability and pharmacokinetics in rabbit eyes were studied.The best prescriptions identified in this article were: atropine sulphate drug concentration of 0.01%,methyl cellulose concentration of 1.5%,polyethylene glycol concentration of 8.0%,and sodium citrate concentration of 3.5%.The gelation time of the atropine sulphate gel formulation of the selected formulation was 30 s,the phase transition temperature was 34°C,and the viscosity range was 10-20 Pa.s.The preparation has a release time of up to 17 h in vitro and has a complete release with a certain sustained release performance.In vivo pharmacokinetics showed that the relative uptake ratio and peak concentration ratio of atropine gel preparation in sclera tissue were greater than that of atropine solution.The targeting efficiency of scleral tissue was greater than that of other non-target organs,indicating that atropine sulfate thermo-sensitive gel has targeting for scleral tissue.
