Papain can be obtained from the latex of papaya plants(papaya)and has been widely used in various industrial productions,mainly in meat tenderization,food,feed,brewing and textile and other industrial production.In order to improve its stability and storage capacity,this paper mainly uses hierarchical pores such as motherwort,polyethylene oxide-polypropylene oxide-polyethylene oxide triblock copolymer(P123),tetraethylorthosilicate and other raw materials.Bioactive glass,combined with AIE luminescent material,was characterized by various means.It was found through experiments that a hierarchical structure with a pore size of about7 nm and a two-dimensional hexagonal height order was obtained,and the AIE chromophore was also successfully connected to a hierarchical bioactive glass.Papain immobilization conditions were studied by single factor and response surface optimization protocols,and relative enzyme activity was tested.Single factor experiments showed that the enzyme activity was very high at a p H of 7.0,a fixed temperature of 30°C,a fixed time of 4 h,and an enzyme amount of 2.0 mg/10.0 mg.The response surface experiment found that the p H was 7.08,the temperature was 28°C,and the amount of enzyme was 1.995 mg.At this time,the relative enzyme activity was highest and the enzyme loading capacity was 46 ± 0.2 mg/g.On this basis,the free enzyme and immobilized enzyme were compared accordingly.It was found that there were some changes in the optimum temperature and p H value of the immobilized enzyme.The optimum p H of the immobilized enzyme increases,and the optimum temperature also increases.The sensitivity of the immobilized enzyme temperature,p H,storage time,etc.decreased,and the quality was improved in allaspects.Immobilized papain has good properties,improved product stability,easy storage,and is conducive to industrial applications and development.Finally,the immobilized amount of enzyme was detected by fluorescence.It was found that the material had a higher immobilizing capacity for the enzyme,and the immobilized amount increased,and its luminescence intensity increased.Therefore,the immobilization of the enzyme can be monitored by fluorescence monitoring method.
