In this study,the technology of preparation isomaltooligosaccharide with acorn starch was optimized,and then the process optimization of liquefaction,saccharification and transglycosylation was investigated,optimizing the refining conditions,finally,the effect of the refined acorn starch isomalto-oligosaccharide on the proliferation of Bifidobacterium and its laxative effect was obtained.The specific results are as follows:(1)Using thermostable a-amylase to liquefy acorn starch,and using Box-Benhnken response surface analysis method to optimize process conditions of liquefying.According to the single factor test results designing center combination experiment,with DE value of 13%as the best liquefaction index,and the quantity of enzyme,liquefying time,liquefying temperature,liquefying pH value and the substrate content as influence factors.The optimal liquefaction conditions were 31.1 min of liquefaction time,94.75? of liquefaction temperature,6.58 of liquefying pH value,13.44 U/g of thermostable a-amylase,at this point,in combination with the production practice,the value of the liquefied liquid DE was 12.91%.(2)Firstly,with DE value as the index,and the quantity of pullulanase,the quantity of glucoamylase,saccharification temperature and saccharification time as influence factors,optimizing process conditions of saccharifying acorn starch liquefying liquor.Then with isomalto-oligosaccharide content(IG2+P+IG3)as the index,with a-transglucosidase dosage and transglycosylation time as influence factors,optimizing process conditions of transglycosylation process.The saccharification and transglycosylation process conditions were optimized to be 25 U/g of pullulanase,130 U/g of glucoamylase,60? of saccharification temperature and 10 h of saccharification time,at this optimal condition,the DE value of the saccharification liquid was predicted to 84.69%;the optimum concentration of a-glucosidase was 1.5 U/g and transglycosylation time was 35 h,under the optimal condition,the total content of isomaltose,panose,and isomaltotriose were 36.27%.(3)The purification of acorn starch isomalto-oligosaccharide included activated carbon decolorization and anion,cation exchange resin desalination.Activated carbon decolorization,in which,the decolorization rate was the index,and the amount of activated carbon,decolorization time and decolorization temperature were influence factors,optimizing process conditions of activated carbon decolorization;Using the 001×7 cation exchange resin and 201×7 anion exchange resin to desalinate isomalto-oligosaccharide by series of anion and cation,with the decolorization rate as the index,studying the decolorization rate of these influence factors which are the volume ratio of anion-cation resin,velocity and pH in different isomalto-oligosaccharide volume.Through the dynamic adsorption,it obtained the optimal conditions of desalination process by comparison of the results.The purification process conditions were optimized to be 1%of amount of activated carbon,40 min of decolorization time,30? of decolorization temperature,1:2 of volume ratio of anion-cation resin,1 BV/h of velocity and 5-7 of pH,the effect of pH on the desalination effect was not significant.The decolorization rate of the best activated carbon condition was 53.01%;Under the optimum condition of anion and cation exchange resin desalination,when the isomalto-oligosaccharide volume was 100 mL,the desalination rate reached 96.93%;The content of(IG2+P+IG3)reached 63.5%.(4)Being used as carbon source substitute for glucose in MRS medium,to do the proliferation test and the comparison of growth curves;Using the mice to establish the constipation model group,blank control group,acorn starch isomalto-oligosaccharide low,medium,and high dose groups,at the same time,doing the experiment of mice defecation and intestinal propulsion.Text results showed that the acorn starch isomalto-oligosaccharide had significant promoting effects on the growth of Bifidobacterium adolescentis.With the increase of acorn starch isomalto-oligosaccharide,the viable count of the probiotics increased.When glucose was substituted by the acorn starch isomalto-oligosaccharide with concentration 1.5%,after 24 h culture,the count of the Bifidobacterium adolescentis was(7.22±0.15)×108 cfu/mL,which had reach to maximum quantity,and acorn starch isomalto-oligosaccharide promoted the growth rates of the Bifidobacterium adolescentis and made its stable growth periods earlier;The acorn starch isomalto-oligosaccharide low,medium,and high dose groups all can improve the function of constipation and defecation,in which,the laxative effect of medium dose group was better.
