Preparation Of Compound Sulfamethoxazole Tablrts Containing Sodium Bicarbonate

Objective: Cotrimoxazole,also known as compound sulfamethoxazole.It is by sulfamethoxazole?sulfamethoxazole,SMZ?and trimethoprim?trimethoprim,TMP?two kinds of the main combination of drugs,is an antibacterial agent.For the treatment of respiratory diseases,urinary tract inflammation,enteritis and skin wound infection.As SMZ in the urine acetylation rate is high,and the solubility is low,it is easier to produce crystallization of urine,hematuria and other adverse reactions.So when taking this product should be more water,keep a lot of urine.If the use of the drug dose of large medication for a long time,in addition to a lot of water,should also take NaHCO₃ tablets to prevent crystallization of urine and other side effects.In order to reduce the burden of drug users and the side effects of this drug,so the experimental study of sodium bicarbonate added to the compound promise to achieve this purpose.But SMZ is sensitive to alkaline conditions,the use of non-toxic ?-cyclodextrin as a carrier.Methods: Sulfamethoxazole inclusion complex was prepared by saturated aqueous solution method,and the standard curve was used to determine the inclusion rate and drug loading of the inclusion compound.Solvent is anhydrous ethanol,the molar ratio of inclusion material is 1: 1,the inclusion temperature is 60 ?,the inclusion time is 2h,the filtration is carried out,the ethanol is washed and dried in vacuum for one day.Inclusion of sulfamethoxazole content and trimethoprim ratio of 5: 1 by adding sodium bicarbonate and other accessories made of tablets.Results: The yield of ?-cyclodextrin inclusion compound inclusion complex prepared by saturated aqueous solution method was 55%.The entrapment efficiency was determined by standard curve method and the encapsulation efficiency was 80%.The inclusion was identified by TLC and infrared spectroscopy.The sulfamethoxazole tablet containing NaHCO₃ was prepared with dextrin as the main auxiliary material.It dissolved rapidly in the dissolution media of 0.1N hydrochloric acid for30 minutes and dissolved completely in 60 minutes.Conclusion:The use of ?-cyclodextrin inclusion SMZ to prepare sulfamethoxazole tablets containing NaHCO₃,to improve the safety of clinical application of compound sulfamethoxazole tablets.
