Because of its excellent biocompatibility,silk can be prepared into various biological materials such as hydrogels.Silk protein-based hydrogels have great potential in drug carriers and cell culture.How to extract soluble silk fibroin from silkworm silk to prepare functional bio-materials quickly andefficiently is a problem that has been plagued.This paper starts with the extraction of silk fibroin and combines microwave-assisted heating technology to explore the preparation process of a new protein-based hydrogel and the new type applications of hydrogels in cell culture.The main research contents of this paper are as follows:(1)Ru(?)-photochemical cross-linking method is used in this paper,which allows two tyrosine residues to be cross-linked into di-tyrosine to obtain a novel silk protein-based hydrogel,and some analysis tests have been used.The characterization of the mechanical properties of the hydrogel was performed.The characterization results showed that the novel protein-based hydrogel has good elasticity and its compressive modulus is 349±64 MPa;In addition,its hydrogel has a porous network structure inside.The average pore size is about 130 um;The most important,the stability and excellent bio-compatibility of this protein-based hydrogel make it have a wide range of tissue engineering potential.(2)Soluble fibroin was obtained using two different heating systems and analyzed by scanning electron microscopy(SEM),gel chromatography(GPC),Viscosity tests,Fourier transform infrared spectroscopy(FTIR)and so on.The test method characterizes the product and explores the effect of two different heating systems on the performance and structure of the silk protein solution and silk fibroin.The characterization results show that the microwave-assisted heating system causes the silk to melt rapidly by directly acting on the large molecular chains inside the silk,and the molecular weight of the silk proteinin the silk solution after dissolution is small and evenly distributed,Besides,the yield of soluble silk fibroin obtained under the microwave-assisted heatingsystem is higher than traditional hydrothermal,without causing the destructionof the internal structure of silk.The most important,microwave-assisted heating system has certain role in the promotion of the internal amino acid repair of silk,Therefore,microwave-assisted heating technology has a positive effect on the extraction of soluble silk.
