Liver cancer is a common disease with high morbidity and mortality.Currently,surgical treatment and chemotherapy are the main methods for clinical treatment of liver cancer.Doxorubicin is a common drug for the treatment of liver cancer,it has good effect on liver cancer,but it also has a series of side effects.To alleviate the side effects of doxorubicin,the use of an adjuvant drug with chemotherapeutic drugs has a clear effect.Selenium is an essential trace element which has a high biological activity and many functions such as anticancer,antioxidation,and improving the body immunity.In order to observe whether the combination of selenium and chemotherapy drugs has the effect of promoting the anticancer effect of doxorubicin and can alleviate the toxic and side effects of doxorubicin.In this experiment,biogenic selenium nanoparticles?bioSeNPs?were selected as a adjunctive drug for doxorubicin.HepG2 tumor xenograft model was established by injecting HepG2 cells into nude mice,divided them into 5 groups randomly:normal group?normal saline?,control group?normal saline?,doxorubicin group?5 mg·kg-11 doxorubicin,once a week?,bioSeNPs group(0.3 mg Se·kg-1 bioSeNPs,once a day),combined group(5 mg·kg-1 doxorubicin,once a week and 0.3 mg Se·kg-1 bioSeNPs,once a day).Measuring the body weight and tumor volume of HepG2-bearing nude mice every 7days,after 21 days,eyelid blood,detecting the activity of alanine aminotransferase?ALT?,aspartate aminotransferase?AST?,lactate dehydrogenase?LDH?,blood urea nitrogen?BUN?,total bilirubin?TBIL?,and creatinine?Crea?;the white blood cell count?WBC?,red blood cell count?RBC?,hemoglobin?HGB?,hematocrit?HCT?,mean volume of red blood cells?MCV?,mean hemoglobin?MCH?,mean hemoglobin concentration?MCHC?,and platelets?PLT?of blood;liver index;the antioxidation indicators of liver:superoxide dismutase?SOD?,glutathione peroxidase?GSH-Px?,catalase?CAT?,and malondialdehyde?MDA?;the liver,heart,kidney,lung,and spleen were made into histopathological examination.Stripped the tumor,weighed wet weight and volume of the tumor,calculated the relative proliferation ratio and tumor inhibition rate.The results show that:First of all,the combination of bioSeNPs and doxorubicin can further inhibit the growth of tumors,and the inhibitory rate of the combined group is greater than that of the doxorubicin group and the bioSeNPs group.The tumor inhibition rates were in the following order:combined group?81.10%?>doxorubicin group?79.91%?>bioSeNPs group?35.74%?.According to the changes in the relative proliferation ratio of the tumor,it can be seen that the tumor growth in the combination group is the slowest and less than 60%,which proves that the combination therapy can inhibit the growth of the tumor.Second,doxorubicin will lead to a decline in the body weight.Compared with the control group,the doxorubicin group could reduce the number of RBC HGB,and WBC.And it can improve the content of serum enzyme AST,ALT and LDH.Also doxorubicin could reduce the level of SOD,CAT,GSH-Px,and increase the content of MDA.But when bioSeNPs combined with doxorubicin it can be concluded that combination therapy can alleviate the side effects of doxorubicin.Last,in histomorphology,the liver of doxorubicin has inflammatory cell infiltration and severe bleeding,and there are no significant changes in the liver of other groups.After the combination of bioSeNPs and doxorubicin,it was found that bleeding between liver cells was significantly improved.Other group has no obvious histomorphological changes.In conclusions,bioSeNPs combined with doxorubicin can promote the anticancer effect of doxorubicin,and can also reduce the weight loss caused,serum enzyme release increased,blood cell damage,and enzyme activity of antioxidation enzymes decreased by doxorubicin in nude mice.In the aspect of histomorphology,bioSeNPs combined with doxorubicin significantly improved the bleeding between liver cells.Proved that the combination of bioSeNPs and doxorubicin can alleviate the side effects of doxorubicin and this mechanism may be that bioSeNPs can improve the antioxidation level and prevent cell membrane from being damaged by free radicals.
