Boletus aereus is a kind of delicious,nutrient-rich wild edible fungus,mainly distributed in Yunnan,Sichuan,Guizhou and other places.Studies have shown that Boletus aereus has a wide range of good biological activity,including anti-oxidation,protection and repair of alcoholic liver injury,blood fat and so on.In this study,we wanted to extract and purify polysaccharide from Boletus aereus with good antioxidant activity,and to explore the composition of Boletus aereus polysaccharide and to study the effects of Boletus aereus polysaccharide on immunocompromised mice and S-180 tumor-bearing mice.1.Isolation,Purification and Structure Analysis of Polysaccharide from Boletus aereus.In this study,the polysaccharides of Boletus aereus were extracted by hot water extraction method,and the crude polysaccharides(CBAP)of Boletus aereus were obtained.After deproteinization using the method of Sevage,the CBAP become the refined polysaccharide from Boletus aereus(RBAP).Purified polysaccharide from Boletus aereus(PPBAP ? and PPBAP ?)was obtained from RBAP by DEAE-Cellulose52 and Sepharose 4B.The components and structure of PPBAP? and PPBAP? were analyzed by UV spectroscopy,infrared spectroscopy and high performance liquid chromatography.The results showed that the purity of PPBAP ? and PPBAP? was 90.10±0.99%and 90.01±0.87%.The results of UV scanning showed that both PPBAP ? and PPBAP ? did not contain nucleic acid and protein.The IR spectra of PPBAP ? and PPBAP ? showed that the polysaccharides has furan ring,pyran ring carbon skeleton and other typical polysaccharide characteristic groups.HPLC analysis showed that polysaccharides from Boletus aereus were polysaccharides derived from the polymerization of glucose,fructose,xylose and a small amount of rhamnose.2.Using RBAP as the experimental material The model of immunosuppressive mice was established by cyclophosphamide,and the immune-related indexes were determined.Mice were randomly divided into normal control group,model group,RBAP dose group and positive control group.Changes in immune organs were judged by Determination of spleen and thymus index.The cellular immune function is detected by carbon particle clearance test.The levels of cytokines IL-2 and IFN-y in serum were measured by Elisa method to detect the cellular immune function in mice.The results showed that the RBAP groups could significantly increase the spleen and thymus index,the carbon clearance index and the levels of IL-2 and IFN-? in the serum of mice in immunosuppressed mice compared with the model group(P<0.05 or P<0.01).This suggests that RBAP has a certain potent effect on immune function in immunosuppressed mice.3.Using RBAP as the test material,the tumor-bearing mouse model was established by subcutaneous injection of S-180 sarcoma cells,and the functional test was carried out.Mice were randomly divided into normal control group,model group,positive drug group and RBAP dose group.The effect of RBAP on the immune function of tumor-bearing mice was measured by measuring the sarcoma size,immune organ index and serum levels of IL-2 and TNF-? in mice.The results showed that RBAP could inhibit the proliferation of sarcoma in tumor-bearing mice and significantly increase the spleen and thymus index and the levels of serum IL-2 and TNF-? in tumor-bearing mice(P<0.05 or P<0.01).This suggests that RBAP has a potent effect on the immune function of S-180 tumor-bearing mice and RBAP may inhibit tumor proliferation by immunomodulation.
