Antrodia cinnamomea(A.cinnamomea)is a kind of precious medicinal edible fungus,which was discovered in Taiwan named "Niu-Chang-Ku".This fungus grows only on the special rotten wood of Cinnamomum kanehirai Hay,a rare species of trees.In the past,people always used it as adjunctive therapy drug for alcohol liver,hepatitis and hepatic carcinoma.A.cinnamomea was found contained 4-acetylantroquinonol B(4-AAQB)that was confirmed had efficient anticancer activities with no side effects.Based on this,production of 4-AAQB from A.cinnamomea by liquid deep fermentation is a fact of great importance.This study explored the culture conditions of the biomass accumulation and high-yield of 4-AAQB for A.cinnamomea,and researched a preliminary study on the process of segmental fermentation.The main works are as follows:(1)Study on formation conditions for A.cinnamomea to form the best,the highest dry weight biomass.The results are as follows:glucose 20 g/L,peptone 10 g/L,KH2PO4 0.75 g/L,MgSO4 1.5 g/L,liquid volume 100 mL/250 mL,temperature 281C,rotational speed 150 rpm.It is concluded that the fifth day seed fermentation results have more advantages.(2)This study explored the culture conditions of the biomass accumulation for A.cinnamomea.The compose of culture medium was as follows:lactose 70 g/L,silkworm pupa powder 40 g/L,corn steep liquor 30 g/L,VB5 0.3 g/L,K2HPO4 0.5 g/L,MgSO4 0.5 g/L,The cultivation conditions were as follows:rotational speed 130 rpm,pH=5,liquid volume 80 mL/250 mL,inoculation amount 10%.In this situation,we get the highest dry weight of mycelia of A.cinnamomea 12 g/L,culture time is 8 days.(3)This study explored the culture conditions of the yield of 4-AAQB for A.cinnamomea.The compose of culture medium was as follows:fructose 55 g/L,peptone 40 g/L,beef paste 30 g/L,VC 0.3 g/L,K2HPO4 0.5 g/L,MgSO40.5 g/L.The cultivation conditions were as follows:rotational speed 170 rpm,pH=7,liquid 'volume 120 mL/250 mL,inoculation amount 25%.In this situation,we get the highest dry weight of mycelia of A.cinnamomea 4.5 g/L,and the yield of 4-AAQB reached 2.2 mg/g.The yield of 4-AAQB increased by 1100%,and the fermentation period shortened to 20 days,and the cycle shortened by 30%.(4)This study explored the most suitable carrier is the fabric of chemical fiber.The results are as follows:the best net area is 5 cm*25 cm,and the best placement way is folds,and the best inoculation amount is 0.6 mL.(5)This study explored the different fermentation methods in 5L fermentor.The results are as follows:the mesh fermentation method can get a higher A.cinnamomea biomass and obtain higher yield of 4-AAQB.(6)This study explored the best.extraction method for 4-AAQB.The results showed that the optimal extraction method for 4-AAQB in A.cinnamomea was.by oscillatory extraction.The experimental results show that the optimal conditions for the extraction were as follows:material ratio 1:20(g/mL),the extraction solvent was 90%ethanol,the extraction speed was 150 rpm,the extraction temperature was 30?,the extraction time was 1.5h.In such a condition of extraction,the extraction rate of 4-AAQB in A.cinnamomea can reach more than 97%at one time.
