Study On Preparation Technology Of Compound Hong Qi Sleep-Promoting Granules

Objective: To select the nourishing yin and yang,coordinating heart-kidney,and nourishing blood to tranquilize mind from famous traditional Chinese medicine clinical prescriptions,combined with the theory of traditional Chinese medicine for people who suffer from insomnia,such as blood deficiency,lack of spirit preservation,yin deficiency and yang excess,and heart-kidney imbalance.Through the extraction process optimization,preparation process and quality standards research,we will develop a granule product that can improve sleep function and be suitable for those who have sleepy problems.Significance: The deep sleep will gradually decrease with age.Developing a health product aim to CNS disorder and the toxic and side effects of western medicines will have great sense to improve sleep and the quality of life for those who suffer from insomnia,and have great market demand and broad prospects.The compound granules contain Radix Hedysari,Spina Date Seed,Radix Paeoniae Alba,Radix Rehmanniae Preparata,Fructus Ligustri Lucidi,Matrimony Vine,Oyster,Polygala etc.The combination of above herbs can improve sleep.Taking into the above considerations,traditional Chinese medicine that meets traditional Chinese medicine theory and has a good clinical basis will be developed into a modern preparation.On the one hand,it will ensure efficacy,and on the other hand,it will be convenient to take.Methods: 1.Prescription herbs were extracted by water.2.Multi-components were used as indicators,orthogonal design test was used to optimize the extraction parameters.And the extracting solution was concentrated and dried under reduced pressure.3.Determination of indicator components: The contents of total saponins and total flavonoids were determined by UV,the content of peonidin was determined by HPLC.4.The granules were prepared by wet granulation compression method.5.High-temperature and high-humidity experiments were conducted to study the stability of the preparation.Results: The optimal extraction process was as following: Extracting 3 times in all,1 hour for each time,10 times amount of water into the the medical materials for the first time,8 times amount of water for the second time and the third time.Dextrin was used as excipients the ratio of dry cream powder and dextrin was 1:1.5,85% ethanol was used to pelletize.Three batches of preparations were prepared and the contents were determined.Then,the stability of the preparations was tested.Under the strong light,the appearance of the samples was indifferent and the validity period was predicted to be 2 years.Conclusion: The optimized extraction process had the high yield rate,and the contents of total flavonoids,total saponins and paeoniflorin were high.Taking into account the fact that there are a large number of polysaccharides with hygroscopicity in the medicinal materials,to prepare suitable granules.Stability study confirmed that the quality of preparations was stable within 2 years.
