For a long time,humans have been engaged in the study of ultraviolet rays,skin and their interactions.Ultraviolet rays include 95% UVA and 5% UVB.It is well known that UV irradiation can cause great damage to the skin,including immunosuppression and oxidative stress,as well as the characteristics of rough skin,wrinkles and pigmentation.With the increasing pursuit of the life,searching for direct and effective methods to prevent and mitigat photoaging is the goal of the cosmetics industry.Hydroxysafflor yellow A is a kind of monochalcone compound,which is a natural active ingredient extracted from safflower.At present,the drug has been widely used to treat cardiovascular and cerebrovascular diseases,and studies have shown that it has the biological activity of anti-inflammation and anti-tumor.However,the role of Hydroxysafflor yellow A in photoaging is rarely reported.This paper aims to study the protective effect and molecular mechanisms of Hydroxysafflor yellow A in the oxidative damage induced by UVB and UVA.In the previous experiments,we screened more than 300 natural plant monomers,and found that the Hydroxysafflor yellow A has obvious cell protective activity.By CCK-8,RT-PCR,western blotting,flow cytometry and other methods to detect,results show that Hydroxysafflor yellow A can effectively remove the ROS caused by UVB radiation in HaCaT cells,reduce the attack on mitochondrial membrane,and then obvious fix mitochondrial membrane potential.By up-regulating the expression of bcl-2,and down-regulating the expression of Bax,the effect of anti-apoptosis was achieved.In addition,the abnormal expression of TNF-?,IL-6,IL-8 caused by UVB irradiation was obvious inhibited by Hydroxysafflor yellow A.And its antiinflammatory effect was achieved by inhibiting the activation of P38 MAPK andJNK.we also found that Hydroxysafflor yellow A has anti-apoptotic,anti-oxidant anti-inflammatory effects in HSF cells induced by UVA irradiation.In addition,UVA causes the loss of collagen in dermal dermis,so we further examined the effect of Hydroxysafflor yellow A on the expression of MMP-1,MMP-3,MMP-9 and TIMP-1in HSF cells.The results show that Hydroxysafflor yellow A has anti-photoaging effect.In animal experiments,we used ultraviolet A and ultraviolet B combined irradiation,the experimental results showed that Hydroxysafflor yellow A can effectively alleviate the erythema formation,peeling and inflammation of nude mice.H&E staining showed that the drug significantly inhibited epidermal hyperplasia caused by UV-irradiation.Western blotting results show that Hydroxysafflor yellow A can effectively inhibit MMPs expression,and reduce the loss of collagen,which was consistent with the results of Masson trichrome staining.In accordance with the results of cell experiments,its anti-ultraviolet activity was also was achieved by inhibiting the activation of P38 MAPK and JNK.Taken together,our results suggest that the Hydroxysafflor yellow A has anti-oxidant,anti-inflammatory,anti-photoaging,anti-apoptotic effects.All results provide the good theoretical research basis for drug as a potential cosmetic ingredients.
