Objective:1)In order to provide enidential support on the extracted technology for hydroxysafflor yellow A,we used the HPLC method to determine the content of hydroxysafflor yellow A in Carthamus tinctorius;2)The technology of extraction was improved to elevate the yields of hydroxysafflor yellow A.Methods:1) Detected the content of hydroxysafflor yellow A in in Carthamu stinctoiusl.by using the HPLC method.methanol-acetonitrile-0.7%H3PO4(26:2:72) as a mobile phase were used.The flow rate was 1.0ml/min,the column temperature was 25℃,and the detective wavelength was 403nm.2)reviewed the influence of the yield of the hydroxysafflor yellow A by using different extracted methods:the methods of clarifier,ethanol precipitation,macroporousresin,clarifier combined macroporousresin,clarifier combined MCI gel CHP-20p;3)Study design monofactor designation and orthogonal designation,A comparative study was performed among four kinds of ablation technologies.Results:1)The method of ZTCⅡclarifier:The purity of HSYA was 90.8%,the yeild was 0.18%;2) The method of ethanol precipitation:The purity of HSYA was 91.3%,the yeild was 0.19%;3) The method of macroporousresin:The purity of HSYA was 94.8%,the yeild was 0.47%;4) The method of clarifier combined macroporousresin:The purity of HSYA was 97.3%,the yeild was 0.46%;5) The method of clarifier combined MCI gel CHP-20p:The purity of HSYA was 93.9%,the yeild was 0.51%.Conclusion:extracting program is as followering:The extracting solvent was water by using ZTCⅡclarifier as the purifying portion at first.The yield and the purity were higher by used D-4020 macroporous resin column chromatography and Sephadex LH-20 chromato-graphy.The technology make clear that the method was cheap and the yield is higher.This technique is suitable to prepare HSYA.
